Studies On The Purification Process And Its Effects Of Deep Partial Burn Wound Healing In Mice Of Polyphenols In Longan Seeds

In order to use this kind of by-product, which is discarded during longan fruit processing, the main chemical components in longan seeds were analyzed. Solvents with different polar, Macroporous adsorption resin and Sephadex LH-20 gel were employed to fractionate and purify polyphenols from longan seeds and the main composition of the polyphenols were analyzed by HPLC and HPLC-ESI-MS. The antioxidant and antimicrobial activities of the different fractions were also inverstigated, meanwhile,explored the effects of rats deep partial burn wound healing of polyphenols in longan seeds.The main results were shown as follows:(1) The results of the main components analysis showed that longan seeds contain abundant starch, reducing sugar, protein, and it can be used as a better nutrition resources.Meanwhile, the content of fiber and fat is relatively less, also it contain some of content polyphenol compounds.(2) The results of liquid-liquid extraction was used to separated polyphenols of longan seeds showed that the ethyl acetate extract had the highest content of total polyphenols,which was 693.51±9.52 mg/g among the four extracts. The four extracts had a relatively weak scavenging effect on superoxide anion radicals but could strongly scavenge ABTS and DPPH and hydroxyl free radicals, and the antioxidant activity of ethyl acetate extract was highest. The four extracts had inhibitory effects of different intensity to Salmonella,Escherichia coli, Staphylococcus aureus, and the ethyl acetate extract was especially outstanding, except for the water residue on the inhibition of Salmonella and Escherichia Coli had no obvious effect.(3) Comparing with the polyphenols' adsorption and desorption rates of seven kinds of macroporous to choose the suitable macroporous resins, the static tests results showed that adsorption and desorption rates of AB-8 were 69.65 and 94.47% respectively was selected as the best macroporous resin. The static and dynamic experments showed that the optimum conditions for column chromatography were: p H of 3.5, polyphenols concentration of 2 mg/m L, and the flow rate of 2 m L/min, the eluant concentration of 70 %ethanol and the elution flow rate of 2 m L/min. The recovery, yields and purity of longan seed polyphenols made under these conditions was 75.65%, 4.93% and 65.84%respectively.(4) The results of polyphenols gradient elution with water-ethanol solution on the AB-8 resin showed that composition and content of polyphenols exist some differences in ethanol eluates with different concentration, indicating that AB-8 resin could be used to preliminarily separate and purify polyphenols in longan seeds. In vitro antioxidant and antimicrobial activities of longan seed polyphenols were studied, the results showed that the activities of 30% ethanol eluate were better than the other ethanol eluates in general.There were five components in the elution curve, which separated by Sephadex LH-20 from 30% ethanol eluate. HPLC and HPLC-ESI-MS analysis showed that the main active constituent of longan seed polyphenols were ellagitannins compounds such as ellagic acid and corilagin.(5) By wound healing rate, determination of biochemical indexes and histopathology analysis to evaluate the quality of wound healing. The results showed that middle and high dose groups had a certain role in promoting to wound contraction in the early stage of burn healing, subsequently the healing rate had no noticeable difference compared with normal saline group at different time points; A dose of longan seed polyphenols solution had a certain effect on the variation in content of protein and hydroxyproline in wound tissue and serum; In inflammatory cells infiltration, hyperplasia of fibroblasts and epithelial cells,formation of capillaries and granulation tissue to evaluate histopathologic, longan seed polyphenols in each dose group compared with normal saline group had no obvious difference. Therefore, longan seed polyphenols have a certain influence to deep partial burn wound healing in mice, but there was no obvious role in promoting.
