Past studies had shown that 18?-GA had the function of prevention and treatment for cancer. However, the reports of research on its molecular mechanisms are few. The previous studies of our research group showed that 18?-GA could inhibit cell proliferation significantly and regulate expression of some genes associated with cancer in HepG2 cells. On the basis of previous research, this study investigated the effect of 18?-GA on the cell apoptosis and the expression of c-jun associated with apoptosis in HepG2 hepatoma cells. The result preliminary revealed effects of 18?-GA on cell apoptosis and the molecular mechanisms in HepG2 hepatoma cells. The results of this study are as following:1. Effect of 18?-GA on induction of cell apoptosis in HepG2 hepatoma cells. HepG2 hepatoma cells were cultured with 18?-GA in the condition of selected dose(60 ?g/mL) and selected time(0 h?6 h?12 h?24 h and 48 h,0 h as control group). Using flow cytometry, we investigated the effects of 18?-GA on induction of cell apoptosis in HepG2 hepatoma cells. The induction rates of cell apoptosis of 6 h group?12 h group?24 h group and 48 h group increased from 10.09% to 31.79%?42.2% and 66.91% compared with 0 h group,and each of the 4 experiment groups differed significant(P?0.05) compared with 0 h group. These results indicated that 18?-GA caused a significant induction of cell apoptosis in HepG2 hepatoma cells in a time-dependent way.2. Effect of 18?-GA on the expression of c-jun in HepG2 hepatoma cells. HepG2 hepatoma cells were cultured with 18?-GA in the condition of selected dose(60 ?g/m L) and selected time(0 h?6 h?12 h?24 h and 48 h,0 h as control group). Using western blotting and real time-PCR, we investigated the m RNA and protein expression of c-jun. When HepG2 hepatoma cells were cultured with 18?-GA in different time(6 h?12 h?24 h and 48 h), the mRNA expression of c-jun were up-regulated by 2.5 times(P?0.05), 2.7 times(P? 0.05), 3.2 times(P? 0.05) and 6.2 times(P? 0.05) compared with control group(0 h group); the protein expression of c-jun were up-regulated by 3.8 times, 6.2 times, 8.4 times(P?0.05) and 17.0 times(P?0.05) respectively. These results showed that 18?-GA caused increased expression of c-jun gene, which are roughly in line with the trend of apoptosis rate. These data suggest that the effect of 18?-GA on cell apoptosis of HepG2 hepatoma cells may related to the increased expression of c-jun gene.
