Subjects: To investigate the pathogenic manner and genomic loci of integration of Epstein-Barr virus(EBV) infecting tissue of adenoidal hypertrophy among children.Methods:We use the next-generation sequencing technology to detected 16 excised adenoidal tissues(11 EBV positive samples are experimental group compared with 5 EBV negative samples). Bioinformatic technology was used to analyze the statistics.Results: 149(average) integration sites(supported reads >=1) and 75(average) integration sites(supported reads >=2) were found in EBV(+) specimens. The coverage of reads was up to 30.31%. Five frequent integration breakpoints in human chromosome were found in EBV(+) specimens, including MORF4L2, BTN2A2, SORT1, VAT1 and PLB1. There is no integration site observed on EBV(-) specimens.Conclusion: Our study found that EBV integrating into human chromosome can lead lymphocytes pathologically proliferating. There are 5 viral integration sites found on EBV infected children adenoids, regulating lipid metabolism, B lymphocyte proliferation, stimulating autoimmune procedure andnearby protein promoting tissue hypertrophy.
