Objective:To detect the methylation level of the C13orf18,TERT and SLIT1 genes of the cervical exfoliated cells from cervical lesion and discuss their relationship with the occurrence and development of cervical cancer.To test them in the value of early diagnosis and screening for cervical cancer.Methods:In the colposcope outpatient to collection the cervical exfoliated cells from the patients who have the first time for taking the cervical biopsy.According the results of the pathological result,the cervical exfoliated cells can be divided into different groups the noemal group is 40,CINI-III the every group is 40,the cervical cancer group is 60. To adopt the DNA extraction kit for extracting DNA samples and bisulfite transformation and test the methylation of C13orf18,TERT,SLIT1 with pyrophosphate sequencing technologies.To compare the methylation level of the three gene in different pathological changes and evaluate them in cervical cancer lesion and the diagnostic value of cervical cancer.The data were analyzed by SPSS 17.0software,using the single factor analusis of variance(One-way ANOVA) to analyze the methylation levels of the three genes and the comparion of two group being by the SNK-q.To draw the receiver-operating characteristic cure(ROC),calculating the AUC value,analysize the best threshold.Results: 1. The C13rof18 gene methylation of the normal group,CINI-III group and cervical cancer group respectively was( 2.17±1.24) % ?( 2.47±1.5) % ?(4.31±2.69)%?(12.33±9.94)%?(33.07±16.48)%,the C13orf18 methylation levels of the five groups were statistically significant(F=46.05,P<0.001)? The TERT gene methylation of the normal group,CINI-III group and cervical cancer group respectively was(6.39±2.30)%?(6.17±2.19)%?(7.89±3.98)%?(7.18±3.97)%?(25.15±14.01)%,the TERT methylation levels of the five groups were statistically significant(F=21.68,P<0.001). The SLIT1 gene methylation of the normal group,CINI-III group and cervical cancer group respectively was(6.32±2.31)%?(6.48±2.58)%?(9.72±4.10)%?(16.20±11.29)%?(28.59±16.30)%,the SLIT1 methylation levels of the five gruos were statistically significant(F=15.52,P<0.001).It can be see that the methylation levels of the three genes were changed with the pathological changing.The methylation levels of normal group was low but of the cervical cancer group were improved significantly.2.The sensitivity and specificity of the C13orf18 gene methylation diagnosising the CINII and above lesios were 67.9%,97.7%,CINIII and above lesions were78.1%,98%.TERT42.9%,96.6%,50%91.3%.SLIT166%,92.6%,71.4%,95.2%.While the sensitivity and specifity of HC2-HPV DNA test were 80.7%,36%,86.7%,23.2%?Although the sensitivity of gene methylation dection was lower than HCII test,but its specifity was significantly higher.3.With drawing the ROC cure, combining the three gene methylation for the dection of cervical cancer,the sensitivity and specifity about CINII and above lesions were 78%,96.9%,CINIII and above lesions were 84.4%,96%.The overall is superior to the signle gene methylation dection.Conclusion:1.The hypermethylation of C13orf18,TERT and SLIT1 exist in cervicalcancer.2.The specifity of C13orf18,TERT and SLIT1 genne methylation decting the cervical cancer and precancerous lesions are high.3.Combining of the C13orf18,TERT and SLIT1 genne methylation dection has potential application value.
