ObjectiveBreast cancer, as the leading cause of women,s cancer-related death worldwide,The diagnosis and treatment becomes effective measurements that can imoroving the survival rates in the early. Current research work shows that PVT1 is highly expressed in both breast cancer cells and malignant breast tissues. Its expression level is closely realted with proliferation and invasive growth, but the mechanism is not fully understood. To explore its mechanism in the development of Breast cancer, we explore the relationship between host gene PVT1 and miR-1207-5p and its influence of cell functions. We hope the experiment would offer certain scientific basic to guid the therapeutic for the breast cancer in clinically.MethodsThe relative expression of miR-1204?miR-1205?miR-1207-3p?miR-1207-5p?miR-1208 in T47 D were detected by RT-qPCR, miR-1207-5p was chosen as research object, The relative expression level of miR-1207-5p was detected by RT-qPCR in 22 cases of Breast cancer tissues. To discover the relationship between host gene PVT1 and miR-1207-5p at tissue and cell level, Reverse transcription polymerase chain reaction(RT-PCR) detection was used to verify the relative expression level of miR-1207-5p and PVT1 in T47D?MCF7?MDA-MB-231. The relative expression of miR-1207-5p was also detected in PVT1-lowexpression cell system and E2-induced T47 D. Proliferation and apoptosis of T47 D cells pre and post transfection induced by miR-1207-5p were analysed by CCK8 assay, plate clone formation assay. Finally,bioinformatics approach was utilitzed for predicting the target genes of miR-1207-5p.The regulatory effect of miR-1207-5p on STAT6 was verified by detecting the expression level of STAT6 in T47 D with miR-1207-5p overexpressed.Results1. The relative expression level of miR-1205?miR-1206?miR-1207-3p in T47 D were undetermined. and, the relative expression level of miR-1207-5p was 54.57 times higher than miR-1204.2. The relative expression of miR-1207-5p in breast cancer is significantly elevated compared to nomal tissues.3. The trend of the relative expression of PVT1 and miR-1207-5p were connsistent4. The relative expression level of miR-1207-5p was decreased in PVT1-lowexpression T47 D compared to control.5. The relative expression level of miR-1207-5p was increased in E2-induced T47 D with higher expression level of PVT16. The increased proliferation of T47 D with overexpression of miR-1207-5p by CCK8 and plate clone formation assay.7. The target genes of miR-1207-5p was predicted by bioinformatics approach, the relative expression level of STAT6 was decreased with the overexpression of PVT1Conclusion1. lnc RNA PVT1 can regulate the expression of miR-1207-5p by the common promoter.2. miR-1207-5p can affect cell proliferation by regulating the expression level of STAT6 directly, and then impact on the development of Breast Cancer...
