Objective: Recently,taking advantages of the mature theoretical system as well as inexpensive,simple,and low time consuming operation,the technology of Capillary Electrophoresis(CE)has been applied on a large mount of researches about the prediagnosis disease.Meanwhile,the Capillary Zone Electrophoresis(CZE)is one of the most general separation model.Following the popluarization of the technique of Mass Spectrum(MS),the CE-MS has becoming a topic that most of global research groups focused on.However,the sophisticated operating process and the high cost jeopardise this method for appling on the clinic detection.This work proposed a method which are based on the semiquantitative method for detecting specific urinary proteomics of patients with Bladder Cancer(BCs)as well as the pathogenic bacteria of urinary infection by Capillary Electrophoresis separation technique coupled with diode array detector(CE-DAD),detecting condition of this method has optimized,preliminary explorating the method for detecting the pathogenic bacteria of usual urinary infection.Methods: For detecting the difference between the contents of each urinary low-abundance proteins from the Bladder Cancer group(BCs,5 patients)and the healthy group(5 volunteers),electrolytic additives have been used to improve the buffer system,and the concentration of electro-osmotic flow is well controlled by this condition,in order to increase the separation efficiency.6 standard strain bacteria have been cultivated and sampling each of them according with the threshold used for diagnosis on the clinic,in order to simulate the concentration of patients' samples,running condition has been optimized,difference between the retention times of each strain has been improved.Results: The optimum condition has been established to detect the difference between the bladder cancer and the control group by CE-DAD,4 of 6 peaks among the experimental and control groups were significantly different according to a t-test(P<0.05),and 6 specific peaks were repeatedly detected in the CIS group,and the difference and correlation of them are also certified by the parallel experiment ofELISA method.Corresponding method has been established to identify the strain of the bacteria,repetitive experiments have also certified at the better resolution,which means that the migration times of each strain have been confirmed under such condition,at the same time,this work has also proposed the gas drive can contribute to separate two strains during the electrophoresis process.Conclusion: This paper makes a preliminary exploration on the condition of difference detecting urinary proteomics and the pathogenic bacteria of urinary by CE-DAD,established the foundation for the institution of standardized experimental method in the future.
