Produce a toxic fungal metabolites frequently, its human, animal and plant have a serious harm, including zearalenone(ZEN)toxin is the most representative, it is mainly in poultry and cereal crops, to give people economic and causing serious loss of life and damage. ZEN toxin research, for discovery and preventive mycotoxins on humans, animals and plants harm is under a very important significance. Human umbilical vein endothelial cells are the organization most representative of the tissue cells, its cellular structure obvious, so often used for various medical research. In this paper, different concentrations of ZEN on HUVEC cells affect the results of further research and analysis of the toxic effects of ZEN. The effects of ZEN on HUVEC cell proliferation, cell viability, cell cycle, oxidative stress, cell secretion, mitochondrial membrane potential, cell apoptosis and the expression of cell cycle- and apoptosis-related genes were detected by using the methods of fluorescence staining, flow cytometry, qRT-PCR, the results were as follows:(1) ZEN can change HUVEC growth morphology of cells to kill the cell structure, impede cell growth and proliferation, and cell viability in serious decline.(2) ZEN can lead to HUVEC cells LDH content gradually increased, indicating that the ZEN highly toxic effect, and the greater the concentration of toxic ZEN stronger.(3) With increasing concentration of ZEN, ZEN on HUVEC oxidative stress will become increasingly strong, can ROS fluorescence enhancement, increased MDA content, reduced GSH content significantly decreased SOD and CAT activity.(4) ZEN can reduce mitochondrial membrane potential HUVEC cells, prompting nuclear staining gradually increased. The nucleus concentrated cell apoptosis.(5) ZEN can HUVEC cellular oxidative stress CAT, GSX-1 expression levels were significantly lower, which also shows the ZEN can destroy HUVEC cellular oxidative stress system.The results showed that ZEN has toxic effects, it can change cell structure, promote oxidative stress, inhibition of cell growth, causing apoptosis. and its toxic strength with increasing concentration increased.
