Objective To test the dipeptidyl peptidase-4 inhibitor-sitagliptin on the T2 DM animal model--ZDF rat. To test the dipeptidyl peptidase-4 inhibitor-sitagliptin on ZDF and SD rat OGTT model.Method The male ZDF rat, 4-6 weeks, will be grouped into three different groups: the vehicle, the pioglitazone and the sitagliptin group. All the animals will be treated once per day via oral gavage for 14 days. An oral glucose tolerance test conducted on day 15. Then after a 10 days recovery period, the second OGTT conducted on the day 25. The animal will be sacrificed after the second OGTT and the body weight, the weight of the liver, kidney, epididymal fat, brown fat, soleus and gastrocnemius weighted to calculate the organ weight index. Collect the blood samples to measure the total cholesterol, triglycerides, free fatty acids and high density lipoprotein C. The food intake and the body weight measured during the study. Forty SD Rats, 6-8 weeks age were grouped for 4 groups: the vehicle, sitagliptin 10, 60 and 100mg/kg. The animals were fasted for overnight, tested the blood glucose as the base line. The sitagliptin and vehicle were administrated at-30 minutes. The glucose(2g/kg) were administrated after sitagliptin and vehicle 30 minutes. Measure the blood glucse at-30, 0, 15, 30, 60 and 120 minutes.Result The sitagliptin can reduce the blood glucose level in ZDF model. The animal body weight treated with sitagliptin had no difference when compared with vehicle group. The animals CHOL, TRIG and HDL-C treated by sitagliptin is lower than control group but no difference. The animal organ weight index treated by sitagliptin had no difference when compared with vehicle group. The animal organ weight index(liver and kidney) treated by pioglitazone had a significant difference when compared with vehicle group. The animal glucose tolerance was improved after treated with sitagliptin and pioglitazone. The acute treated with sitagliptin on SD rats can reduce both the peak and the AUC during the OGTT.Conclusion Sitagliptin can reduce the ZDF rat blood glucose significantly, and can improve the glucose tolerance as well. Unlike the pioglitazone, sitagliptin will not effect the animal's body weight gain. The sitagliptin can reduce the CHOL, TRIG and HDL-C level but no significant difference. There is no effect on body weight index. The sitagliptin can improve the SD rat glucose tolerance during the acute OGTT.
