Objective:This experiment chooses two kinds of metal ion Cu2+ and Fe3+,Add it in the hydrogel?RADA16-NBD?3D cultivated of osteoblast?MC3T3-E1?,Through the corresponding test methods.Observe and compare the two Metal ions on osteoblast proliferation and osteoblast differentiation related effects.Lay a foundation for subsequent mechanism studies and animal experiments.Methods :Put the osteoblast vaccination in RADA16-NBD hydrogel to vitro threedimensional cultivation.Divided into three groups of experiments:control group A and experimental group B(Cu2+)and group C(Fe3+).Cultivate 24,48,72 h,CCK-8 method to detect cell proliferation;cultivate 1,3,5d,ALP detection cell activity of alkaline phosphatase;Induced cultivate 21 d,observe the cell calcified nodules;Transwill chamber cultivate 24 h,ability to observe cell migration.Results:Compared with the control group,There are different degree increased the experimental group osteoblast proliferation ability,alkaline phosphatase activity,the mineralized matrix deposition and cell migration ability have different degree increased.And group B(Cu2+)effect is better than that of group C(Fe3+).The difference has statistically significant?P<0.05?Conclusions:Cu2+and Fe3+can promote the osteoblast proliferation,differentiation and migration also Cu2+effect is superior to Fe3+.
