Procalcitonin(PCT) is the pre-hormone of calcitonin, which is normally secreted by the C cells of the thyroid in response to hypercalcemia. Serum PCT is useful in predicting bacteraemia and in assessing disease severity in patients. Therefore, the studies and in vitro diagnostic method of PCT in clinical medicine have caused wide scholars concern over the recent years. Setting up the colloidal gold semiquantitative immunochromatographic assay for rapid detection of PCT and the enzyme-linked immunosorbent assay detection method of PCT. Optimizing the reaction conditions, the experimental consumables, the reagents to achieve the best detection results by experiments. We also evaluated the sensitivity?repeatability and specificity of detection reagent. Experimental results showed that the colloidal gold semiquantitative immunochromatography assay for rapid detection of PCT has been successfully established, and we also maked the standard color card. The detection limits are10 ng/ml?2 ng/ml?0.5 ng/ml?0.1 ng/ml, have good color developing effect and sensitivity.Performance evaluation showed that the detection reagent had good specificity?repeatability and thermal stability. Compared with domestic kit,it has better color gradient, test line and the results. Successfully established the enzyme- linked immunosorbent assay det- ection method. Determined the critical value of the positive samples is OD450nm=0.2395, the linear range is 75 ng/L-1200 ng/L,the minimum detectability is 75 ng/L. Performance evaluation showed that the detection reagent have good specificity, the coefficient of variation of intraassay and inter-assay was less than 7%, the detection reagents are capable of very high precision.Compared with domestic kit,it has better sensitivity, linear relationship.
