With the development of technology, the method to treat the cancer has a rapid development, anticancer drugs and gene therapy are the hot area of research.There are two purposes in this paper, a simple and low toxicity of polymer drug carriers was develeped,and used it for pulmonary drug delivery, which provided a new kind of simple treatment for the lung cancer. On the other hand, a kind of biodegradable polymer gene carrier was prepareed, and the performance of this polymer was evaluated through the experiment in vitro characterization of the cells, and a reference of high polymer gene carrier for gene therapy was provided. The main contents are as follows:Through the electrostic interaction to combine ?-PGA with DOX, drug delivery system of ?-PGA-DOX was obtained. The particles size of materials was about 100 nm through TEM, the experiment of drug release showed the ?-PGA-DOX had a good release effect under acid condition, the MTT showed the ?-PGA was non-toxic, but ?-PGA-DOX had a good capacity to kill the cancer cells, flow cytometry and CLSM illustrate the cells had the good effect of endocytosis for ?-PGA-DOX, and durg distribution experiments confirmed pulmonary drug delivery way had low toxicity to other organs.Using m PEG-NH2 as initiator, prepared the m PEG-PLL-PPhe and m PEG-PLL-PAla through the ring opening polymerization reaction as gene carrier. TEM and Zeta-Pals illustrated that the materials were nano-scale, zeta-potential showed the m PEG-PLL-PPhe and m PEG-PLL-PAla were positively charge, had the ability to combine with the negatively charged gene. By compared with the PEI called gene transfection gold standard, the following results were concluded: Transfection experiment showed that the material had low DNA transfection, but in si RNA silence experiment, the materials had better effect than PEI. MTT experiments had the lower toxicity than PEI in various cells, the results showed the materials had the research prospect as si RNA gene carrier for gene transfection.
