Role Of Preconditioning With Respectively Honokiol/Baicalin In Protecting Kidney Against Ischemia-reperfusion Injury In Rats

Objectives In this experiment, preconditioning with respectively honokiol/baicalin was evaluated to have protective effects in several renal I/R injury models which was induced in male Sprague Dawley rats by placing a clamp across left renal artery for 40 min following a right nephrectomy.Methods 40 male adult SD rats following randomly were respectively divided into four groups: Sham group, IR group, I/R + preconditioning with honokiol( HNK-Pre group) and I/R + preconditioning with baiclin( BA-Pre group). In the sham control group, rats were subjected to right nephrectomy and a left flank incision without occluding renal vessels. The rats in the I/R groups were subjected to right nephrectomy through a right flank incision, while an additional left flank incision was made to occlude the left renal artery and vein with a non-traumatic vascular clamp for 40 min. For the HNK-Pre group, the same surgical procedure as in the IR group was performed, and honokiol( 50?g/kg) was administrated at the 15 th min before ischemia. For the BA-Pre group, the same surgical procedure as in the IR group was performed, and baiclin(100mg/kg) was administrated at the 15 th min before ischemia. At the end of the ischemic period, the clamp was released for blood reperfusion for 6 h. At the end of reperfusion for 6 h, then the rats were sacrificed, blood samples were drawn for the detection of the renal functions assayed by BUN and SCr, and the left kidney was processed for the detection of renal tissue.Results 1 Compared with the sham group,the contents of Scr, BUN, MPO, MDA, Paller,s score and Cyt-C were significantly increased,but the content of GSH-Px was deceased in the HNK-Pre group, I /R group and BA-Pre group, the significance is not statistical. 2 Compared with the ischemia-reperfusion group,the contents of Scr,BUN, MPO,MDA,Paller,s score and Cyt C were significantly decreased,but the content of GSH-Px was increased in the HNK-Pre group and BA-Pre group(P<0.05). 3 Compared with the BA-Pre group,the difference from the contents of Scr, BUN, MDA and GSH-Px in the HNK-Pre group was not statistical. 4 Compared with the BA-Pre group,the contents of MPO, Cyt-C and Paller,s score were significantly decreased in the HNK-Pre group, therefore the difference was statistically significant.Conclusions The data reveals that honokiol/ baiclin may have a significant protective effect on renal ischemia reperfusion injury through inhibiting oxidative stress and reducing inflammatory reactions. Honokiol/ baiclin,moreover, can reduce the production of cytochrome C which may explain that honokiol/ baiclin can refrain the mitochondria transformation hole open and lessen the discharge of Cyt-C, and can inhibit the mitochondrial apoptosis pathway and alleviate the effect of renal ischemia-reperfusion injury,honokiol took the more significant advantage in the anti-inflammatory property and in the anti-apoptotic property by inhibiting the he mitochondrial apoptosis pathway than baiclin.
