Objectives Three-dimensional reconstruction of the maxillary second premolar by using micro-CT and Mimics10.01 software and analyze its the root canal morphology.Methods 120 maxillary second premolars were scanned by Micro-CT, the incidence of canal isthmus and root canal morphology was analyzed after 3D reconstruction.Results In the 120 single rooted maxillary second premolars, the ?types of root canal morphology have51, which account for 42.50% of the total quantity, the ?~? types of root canal morphology have 69, which account for 57.50% of the total quantity. The incidence rate of canal isthmus is 93.75% in the 69 samples what root canal morphology were ?~? types. The incidence of canal isthmus in the 120 maxillary second premolars is 47.62%, the rate of sheet connections type and mixed type in the single rooted maxillary second premolars what have the canal isthmus are 82.70%,the apical 4~5mm region of the root canal with isthmus have the highest incidence in all sections.Conclusions Micro-computed tomography technology is an effective method to study the root canal system, it is complex that root canal system of the maxillary second premolar,the apical 4~5mm region of the root canal with isthmus have the highest incidence in all sections, if we can reinforce the cleaning and filling of canal isthmus in the 4~5mm section of the apical, the success rates of root canal therapy and root-end surgery will be improved.
