Study Of The Relationship Between Expression Of Galectin-3 In Window Stage Of Endometrial Implantation And IVF Pregnancy Outcome

Objectives To investigate the relationship between the expression of Galectin-3 in window stage of endometrial implantation and the IVF(in vitro fertilization and embryo transfer)pregnancy outcome.Methods Patients with IVF treatment in our hospital from Jan.2014 to Dec. 2015 were examined by transvaginal sonography for endometrial thickness and morphology a cycle before IVF treatment when the average size of the follicles were about 1.8~2.0 cm in diameter. And the endometrial biopsies were taken on day 6 or 7 after ovulation. RTPCR and Western Blot were used to test the expression of Galectin-3 m RNA and protein, in order to evaluate the relationship between Galectin-3 express level and pregnancy outcome. Among all the 378 patients with IVF, there were 45 with biochemical pregnancy, 34 with spontaneous abortion after clinical pregnancy and 141 normal birth. Combined with clinical data, statistical analysis of the experimental data was used to evaluate the relationship between Galectin-3 expreess and pregnancy outcome.Results 1 The comparison of general condition and therapeutic condition in the three groups, such as ages, years of infertility, type of infertility, male factors, body weight, body mass index, basic FSH, sinus follicle number,days of Gn, the dosage of Gn, endometrial thickness on the day of transplantation, the number of transplated embryo and the number of the capable embryos. There were no statistical differences(P>0.05). 2 The comparison of endometrial thickness and morphology in the three groups: There's no difference between three groups in endometrial thickness and morphology.(P>0.05) 3 The expression of Galectin-3 m RNA in endometria in three groups:.There's statistic difference between the spontaneous abortion after clinical pregnancy group(0.18±0.01, n=34)and the normal birth group(0.65±0.03, n=141)(P <0.05). 4 The expression of Galectin-3 protein in endometria in three groups: There's significant difference between the spontaneous abortion after clinical pregnancy group and the normal birth group(P <0.05).Conclusions 1 There were no significant differences about general condition and therapeutic condition among three groups. 2. There were no significant differences about endometrial thickness and morphology among three groups. 3. The expression of Galectin-3 m RNA for normal birth goup was higher than the abortion group in window stage of endometrial implantation4 The expression of Galectin-3 protein for normal birth goup was higher than the abortion group in window stage of endometrial implantation 4. Higher expression of Galectin-3 in window stage of endometrial implantation predicted positive pregnancy outcome,and the endometrial thickness and morphology cannot serve as predictors of positive outcome of pregnancy.
