Objective:To define the ischem-reperfusion injury associated with mitophagy; To research the role of mitophagy which associated with Drp1 in IPC.Methods : Simulated ischemia reperfusion with SD mice. The SD mice were randomly divided into four groups:shame group,I/R group,IPC group,P110+I/Rgroup.The expression of Drp1 and P62/SQSTM by Western blot.Reactive oxygen species(ROS) levels,ATP levels,LDH levels were detected by ROS assay lit,ATP assay kit,LDH assay kit.Results:(1)Compared with Shame group, the activity of ROS in I/R group were significantly higher(P<0.01), Compared with I/R group, the activity of ROS in IPC group were significantly ruduced(P<0.01),Compared with IPC group, the activity of ROS in P110+IPC group were significantly reduced(P<0.01)(2) Compared with Shame group, the concentration of ATP in I/R group were significantly reduced(P<0.01), Compared with I/R group, the concentration of ATP in IPC group were significantly higher(P<0.01), Compared with I/R group, the concentration of ATP in P110+IPC group were significantly higher(P<0.01)(3) Compared with Shame group, the concentration of P62 in I/R group were significantly reduced( P<0.01), Compared with I/R group, the concentration of P62 in IPC group and P110+I/Rgroup were significantly higher(P<0.01).(4) Compared with Shame group, the concentration of Parkin in I/R group were reduced,but P<0.01, Compared with I/R group, the concentration of Parkin in IPC group and P110+I/R group were reduced P<0.01.(5) Compared with Shame group, the concentration of Drp1 in I/R group were significantly higher(P<0.01); Compared with I/R group, the concentration of Drp1 in IPC group and P110+I/R group were significantly reduced.Conclusion:Ischemia preconditioning(IPC) and inhibit mitochondrial division can renduce myocardialreperfusion injury.The mechanism of ischemic preconditioning cardialprotecyion is that,it can inhibit unselective autophagy but not the selective autophagy.
