Objective: To explore the association between serum levels of Matrix Metalloproteinase-2/9(MMP-2/9) and Alzheimer,s Disease(AD) as well as amnestic mild cognitive impairment(a MCI).Methods: The serum levels of MMP-2/9 were measured by enzyme-linked immuno sorbent assay(ELISA) in 40 controls, 44 amanestic mild cognitive impairment(a MCI) and 48Alzheimer's disease(AD).Result: The levels of serum MMP-2 in both AD and a MCI group were higher significantly compared with the control group. Additionally the level of serum MMP-9 in AD group is higher than both the a MCI and control group. Logistic regression analysis showed that the serum levels of both MMP-2(OR 1.217,95% CI 1.072~1.381;P=0.002) and MMP-9(OR 1.102,95% CI 1.007~1.206;P=0.034)were risk factors for AD, and the serum level of MMP-2 was a risk factor for a MCI but not for AD. The serum levels of both MMP-2 and MMP-9 were negatively related to MMSE score, immediate memory score, computation, defer memory score and linguistic capacity, which were found by Spearson correlation analysis. Finally, the results showed that the serum level of MMP-9 but not MMP-2 was negatively related to orientation.The receiver operating characteristic curve(ROC) shows MMP-2 related area under the curve(AUC) is 0.721,and MMP-9 related AUC is 0.763?Conclusions: The serum levels of MMP-2 and MMP-9 were both risk factors for AD, and the serum level of MMP-2 was a risk factor for a MCI. Althoug both the serumlevels of MMP- 2 and MMP- 9 have a certain sensitivity and specificity for the fdiagnosis of AD, they still can not be used as reference factors.
