Objective: To investigate the relative expression of lnc RNA in the plasma from patients of liver cirrhosis, and its correlation with clinical features, aim at providing a new idea for the diagnosis and treatment of liver cirrhosis.Methods: First, all research objects were divided into groups of healthy controls, liver cirrhosis and hepatoma, gathered specimens for quantitative analysis of plasma lnc RNAs that differentially expressed in patients with liver cancer using real-time PCR based method, compared the results between groups to find the target lnc RNAs that differentially expressed between groups of liver cirrhosis and healthy control, as well as groups of liver cirrhosis and hepatoma, the results turned out to be uc002 mbe.2 and lnc RNA-LET. Then, to expand the sample size, we collected blood samples of 23 cirrhosis patients and 20 healthy controls, 6 hepatoma patients for quantitative analysis of plasma uc002 mbe.2 and lnc RNA-LET using real-time PCR. Finally, we maked a comparison between groups using the statistical software SPSS, and analysed the relationship between expression level and clinical features of uc002 mbe.2 as well as lnc RNA-LET.Results: Cirrhosis patients exhibited statistically significantly higher plasma uc002 mbe.2 expression level than healthy controls(P<0.05), while there were no statistically differences between groups of hepatoma and healthy controls, as well as groups of cirrhosis and hepatoma, however, cirrhosis patients exhibited higher plasma uc002 mbe.2 expression level than patients with liver cancer.Hepatoma patients exhibited significantly higher plasma lnc RNA-LET expression level than healthy controls and cirrhosis patients(P<0.05), but showed no statistical difference between groups of cirrhosis and healthy controls. Patients were divied into three groups according to the Child-Turcotte-Pugh classfication criteria, but there were no significant differences between different CTP classifications on uc002 mbe.2 and lnc RNA-LET expression level. Spearman correlation analysis showed that there was no significant correlation between plasma uc002 mbe.2 expression and clinical features of cirrhosis patients such as age, gender, hemorrhage, ascites, biochemical indices, etiology, CTP classification and MELD score, while the expression of plasma lnc RNA-LET of hepatoma patients had a strong negative correlation with age, but no significant correlation with other clinical features.Conclusion: Plasma uc002 mbe.2 expression of cirrhosis is higher than healthy control, as well as hepatoma, and did not affect by common clinical factors, indicate that uc002 mbe.2 may be a potential marker for the diagnosis of liver cirrhosis, but its mechanism needs further research. Hepatoma patients exhibited significantly high plasma lnc RNA-LET expression level, while related literature reported that lnc RNA-LET showed low expression level in cancerous tissue. The specific molecular mechanism of lnc RNA-LET is not clear, and few relevant literature is reported, therefore further study on the specific mechanism of lnc RNA-LET is needed.
