Objectives To observe the therapeutic effects of human Adipose Derived Stem Cells on experimental autoimmune uveitis(EAU) and exploring the possible mechanism of Th 1 /Th 17/Treg lymphocytes and their cytokine production.Method 1.h ADSCs were harvested from liposuction aspirates by adherent culture.The phenotype was characterized by flow cytometry. Induce them into adiposed and osteoblastic differentiation.2. The retinal S antigen emulsified in complete Freund's adjuvant injected in footpads of the lewis rats on day1 and day7 to establish the animal model of EAU. The immunized rats were treated intravenously with h ADSCs on day 12 for consecutive 3 days. In EAU control group,the rats were treated intravenously with an equal PBS. In blank control group, the rats were not treated.On day 15,the eyeball and spleens were colleceted. 3.The inflamemation situateions of EAU were examined and scored according to Caspi.4. We make paraffin slices of rat eyeball exemplar to observe the pathological manifestations and detect IFN-? and IL-17 expression in ocular tissues by immunohistochemistry.5.The Mononuclear cells were obtained from the spleens. We examined them by flow cytometery to evaluate the proportion of Th1?Th17 and Tregs to T cells.Results 1.hADSCs were successfullyseparated and identified. 2.The animal model of EAU was successfully established. Clinical score showed that the EAU rats began to appear inflammation for the first time on day9, reach the peak on day12, then gradually fade. The clinical score of h ADSCs treatment group was lower than that in control group, the difference was statistically significant.(P<0.05).3. Histological examination of the retinal showed significantly less retinal architecturedamage and less inflammatory infiltrates.in h ADSCs treatment group.(p<0.05). 4.Immunohistochemical examination of IFN-??IL-17 expression showed obviously decreased in h ADSCs treatment group.(p<0.05). 5.Flow cytometery : Compare to the blank control group, The proportion of Th1?Th17?Treg were increased in the EAU control group(Th1:15.98±2.39?Th17:12.33±0.622?Treg:2.79±0.14,P<0.05). While the Th1 and Th17 were obviously decreased(Th1:4.11±0.34?Th17:4.34±0.39P<0.05), Tregs were increased larger in h ADSCs treated group(4.79±0.12,P<0.05).Conclusionh ADSCs can effectively prevent and ameliorate EAU, h ADSCsinhibited Th1 and Th17 cells differentiateon and the productions of their hallmark cytokines IFN-? and IL-17,meanwhile,it can promote Treg cells differentiateon.
