| Objectives(1)To study the difference of pharmacokinetic characteristics of eugenol between giving yak heart Baweichenxiangsan and pig heart Baweichenxiangsan in rat;(2)To determine the content of amino acids in the freeze-dried yak heart tissue. Methods(1) 3,4-dimethoxystyrene was used as the internal standard in the detection of eugenol concentration in rat plasma with HPLC and Chromatographic separation was achieved on a ALLtima HP C18(250 mm×4.6 mm,5 μm) chromatography column,using gradient elution with the mobile phase consisting of acetonitrile and water,The flow rate was1.0 mL.min-1,The detection wavelength was set at 280 nm and the column temperature was 35℃. After intragastric administration of the different animal’s heart Ba Wei Chen Xiang San in rats,using DAS 2.0software conducted compartment model fit of average plasma concentration- time data of eugenol to rats.(2)By using the Amino Acid Analyzer to detect amino acids in the cardiac tissue of Yak with ninhydrin post-column derivatization.Results(1) After intragastric administration of rats,that the two-compartment model(weight 1/cc) calculated the concentration of plasma was completely consistent with the measured value,and was superior to other models;The main pharmacokinetic parameters of yak’s heart and pig’s heart Baweichenxiang respectively are that eugenol maximum peak concentration in rats(Cmax)were16.213±0.932 μg·mL-1 and 12.503±1.705 μg·mL-1 、 peak time(Tmax) was 21.667±4.082 min and19.167±2.041 min 、 half-life of distribution(T1/2α)were 20.031±2.963 min and 42.344±19.084 min and half-life elimination(T1/2β)were 69.315±0.000 min.(2)In the test concentration range,detecting 17 types of amino acid, the linear equations correlation coefficient(r) of these 17 types of amino acid was all more than0.9990,the content of the glycine tyrosine phenylalanine arginine amino acid of closely associated with cardiovascular disease accounted for about 12 percent. The rate of average recovery on Glycine, tyrosine,phenylalanine, arginine(n = 6) were 103.2%, 97.7%, 96.9%, 96.4%, respectively, and meanwhile, RSD were all less than 2.0%.Conclusions Two different matching animal cardiac Ba Wei Chen Xiang San’s metabolism of eugenol in rat is not the same. Compatibility of yak heart formula better metabolism, its maximum plasma concentration is higher, faster transfer rate in the body, the amount of absorbed into theblood circulation is more; Yak heart glycine, tyrosine, phenylalanine, arginine content accounted for about12% of the total amount of amino acids, closely related to the cardiovascular disease.it provides the theory basis fromthe amino acid level for yak heart medicine. |
