Research Of Huan Shaodan's Anti-aging Effect On Female And Male Aged Rats

The traditional Chinese medicine(TCM) has the special effects in human anti-aging according to the theory system. Some papers have published for the anti-aging effects of anti-aging prescriptions by TCM. It is suggestion that TCM have the effects beyond the man-made drugs so as to be reused. However, these studies on the antioxidant and the anti-aging effects of TCM are still in the primary stage today and go further to be investigated deeply. This present takes for examply of Huan Shaodan(Let life to return to be young HSD) to test HSD's effects on anti-oxidation and anti-aging at cellular and animal level, respectively.Experiments at the cellular level: firtly, 140 SPF SD rats at the age of 4 months were used to preparing HSD-containg serum and blank serum, respectively; secondly, the HSD-containg serum, blank serum, fetal bovine serum(FBS), HSD-containg serum with VC, FBS with VC were used to prepare different medium, which were employed to culture Rat osteoblast(ROBs); and finally, the proliferation, cycle and expression of mammalian target of rapamyein(m TOR) in ROBs, which cultured at different kinds of medium, were detected to explore the effect of HSD on ROBs.Experiments at the animal level: 28 SPF SD rats(half female and male) at the age of 20 months were randomly divided into HSD female/male groups(FH/MH) and control female/male groups(FC/MC). FH/MH rats were given by gavage HSD once a day for 60 days, while FC/MC rats were given by gavage sterile water once a day for 60 days. The weight each rat is weighed every 7 days during those days. To the last gavage administration the dosages are double, and then the anesthesia of the rats with sodium pentobarbital was injected by abdominal cavity after 1 h and while depth of anesthesia did the following experiments: 1drawing blood from the heart for preparing the HSD-containg serum and blank serum to detect the activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA); 2opening the chest, abdominal cavity and cranial cavity in sequence, removing the heart, liver, spleen, lung, kidney, stomach, pancreas and brain to ice normal saline, respectively, then drying saline with filter paper and weighing organs to calculate the organ index; 3taking 0.1 g(n=5) liver and brain with the addtion of 10%(W:V)physiological saline to prepare liver and brain homogenate by ultrasonication, and then detecting the activity of SOD and the content of MDA, respectively; 4putting brain, heart, liver, spleen, stomach and kidney into 4% formaldehyde to prepare paraffin slice, then dyeing by hematoxylin-eosin(HE) staining in turn. Finally, the slice was observed under optic microscope.The experimental results indicated that at cellular level HSD can promote the proliferation of ROBs, increas the expression of m TOR and regulate cell cycle. However, at the animal level, HSD has no obvious influence on rats' weight, given the guarantee for receiving the futher datas. Compared with female groups, the HSD can significantly slow down the decrease of organ indexes in male rats groups. The activity of SOD had prominent increased in serum, liver and brain, and the content of MDA decreased clearly in male HSD groups. On the contrary, there was no obvious effect of SOD activity in female HSD group, and the reduction of MDA can be observed only in serum. Moreover, the results of HE staining suggested that HSD be potential to keep the organs at stage of young.In summary, HSD has the effects of antioxidant and anti-aging, importantly there are differences between male and famle rats. This mearns that the gender should be considered in studies and usage of TCM.
