Lespedeza cuneata(Dum.Cour.) G. Don belongs to the family Leguminosae, genus Lespedeza. Lespedeza is a large genus of Leguminosae Papilionoideae plants, the genus has about 60 species all over the world. In our country, L. cuneata is mainly distributed in the mountain, wasteland and roadside. The whole grass can be used as medicine. It had been reported in "Shen Nong's herbal classic" that this plant has antitussive, antiasthmatic and expectorant, antibacterial, and promoting diuresis effect. The aerial parts of L. cuneata was isolated and purified using chromatographic methods(Sephadex LH-20, column chromatography of silica gel, MCI gel, preparative TLC, and preparative HPLC) from the portions of petroleum ether, dichloromethane parts, and ethyl acetate. Their structures were identified by spectroscopic methods(ESI-MS, 1H-NMR, 13C-NMR, DEPT, and 2D-NMR spectrum). 12 lignans compounds were isolated from L. cuneata. All the 12 compounds were isolated from this plant for the first time, also isolated from this genus for the first time. 12 compounds isolated from this plant were evaluated for their antibacterial activity against nine bacteria(Riemerella anatipestifer CH1?Bacillus subtilis ATCC6633?Streptococcus suis ATCC43765?Vibrio alginolyticus ATCC17749?Pseudomonas aeruginosa CI 1011?Aeromonas hydrophila CI 1004 ? Salmonella gallinarum CI0812 ?Streptococcus agalactiae ATCC13813 ? and Enterococcus faecalis ATCC19433) using microdilution method. JTL-8 exhibited weak antibacterial activities against Streptococcus suis ATCC43765.
