[Objective] To investigate the expression and promoter methylation of guanine nucleotide binding protein(GNA 12) in preeclampsia.[Methods] The methods of methylation-specific PCR and real-time fluorescence quantitative PCR were applied to examine the methylation of GNA12, mRNA and protein expression,both in 50 cases of placenta tissues and 25 cases of peripheral blood tissues from preeclampsia women and normal pregnant women.[Results] GNA12 methylation extent was lower in preeclampsia group than in controls in the placenta(P=0.003 and 4.3*10-3).GNA12 methylation extent was lower in obesity group than in controls(P=0.04). GNA12 methylation extent was lower in positive group than in negative group with 24 hours urinary protein quantitative(P=0.04).In addition,its mRNA and protein expression level is higher than the controls.Through test of mothers' peripheral blood, the results are similar to the placenta results.[Conclusion] The methylation of GNA12 promoter was associated with the risk of preeclampsia.By detecting the mother peripheral blood, may be able to predict the occurrence of preeclampsia.That is to say,GNA12 can serve as a biomarker of preeclampsia.Metabolic syndrome may be an important cause for preeclampsia.
