Aims: The relationship between HBV replication and the host cell epigenetic factors is a new hotspot. The present study aimed to investigate the relationship between the deacetylase gene h MOF and HBV replication in chronic hepatitis B patients.Methods: 1. Construct lentiviral RNAi library of acetyltransferase gene h MOF. Eight pairs with guaranteed efficiency of sh RNA interference sequence were selected to constrct the lentiviral vectors. 2. Hep G2.2.15 cells were infected with sh RNA lentiviral. q PCR was used to detect HBV pg RNA to conform the interference of efficiency after 72 hours of infection 3. ELISA was used to detect the expression of HBs Ag and Hbe Ag.q PCR was used to detect the level of HBV-DNA after h MOF was silenced 72 hours and 96 hours. 4. The expression of h MOF in the liver of CHB with HBe Ag+ and HBe Ag- were detected at the m RNA and protein to analysis the correlation with HBs Ag, HBe Ag and HBV-DNA.Results: 1. Eight lentiviral vectors of h MOF were successfully constructed. 2. The expression of HBs Ag, HBe Ag and HBV-DNA were significantly deceased after the h MOF gene was silenced.3. The expression of h MOF both with protein and m RNA levels in HBe Ag+ CHB patients, liver tissues were significantly higher than that in control group(P<0.001, P=0.004). The protein level of h MOF were positively corrected with the level of HBs Ag, HBe Ag and HBV-DNA(r2=0.3275,P<0.001; r2=0.22,P=0.0051; r2=0.4195,P < 0.001). For the more, the protein expression of h MOF was significantly higher than the control group in the CHB patients' liver tissues with HBe Ag-negative(P=0.0083) and was positively corrected with the level of HBV-DNA(r2=0.1896,P=0.0377).Conclusion: The expression of HBs Ag, HBe Ag and HBV-DNA were significantly reduced in Hep G2.2.15 cells after silence of h MOF gene. The protein expression of h MOF in CHB patients' liver tissues was positively corrected with level of HBV-DNA. HMOF expression levels of m RNA and protein in the CHB patients' liver tissues with HBe Ag-positive were positively corrected with the level of Hbs Ag and HBe Ag. These data suggest h MOF play an important role in the process of HBV replication.
