Objective: to investigate the mechanism of autophagy induced by PM2.5 on the microvasc?Lar endothelial cellsMethod: Cultured H5 V was stimulated by PM2.5 with different concentrations for 12 hours. Real-time PCR was used to measure the expression of Atg5?Beclin-1?Caspase-3? Caspase-9 m RNA;western blot was used to measure the expression of protein Bax?Bcl-2?LC3B??LC3B??Caspase-3? Caspase-9. high-powered microscope was used to observe the morphology of cells. Transmission Electron Microscope(TEM) was used to observe the autophagosome.Result: The ratio of autophagy associated protein LC3 B ? / LC3 B ? in 100?g/m L group was significantly higher than that in control group. PM2.5(100?g/m L)-induced protein expression of apoptosis associated gene Bax?Caspase-3? Caspase-9 was higher compared with those of control group. PM2.5(100?g/m L)-induced m RNA expression of autophagy associated gene Atg5?Beclin1 was higher than that in control group. PM2.5(100?g/m L)-induced m RNA expression of apoptosis associated gene Caspase-3 ? Caspase-9 was higher than that in control group. PM2.5(100?g/m L)-induced vacuolus were also observed in vascular endothelial cells in which the autophagosomes formed and the amount of apoptotic cells increased.Conclusion: PM2.5 induced autophagy and autophagy promoted apoptosis. |
