Objective:To investigate the expression of Hoxa11 and Wnt/?-catenin signaling pathway in human periimplantation endometrium and explore the relationship of them in regulating endometrial receptivity?Methods: 1.We examined the Hoxa11 and ?-catenin expression in pregnant D1-D5 mice`s endometrial stromal cells at the site of the blastocyst implantation by immunohistochemistry. 2.THESCs was infected by lenti/Hoxa11-sh RN A and lenti/N-control after in vitro decidualization. Then we detected the expression of Wnt4 and ?-catenin through Realtime PCR and Western blot on the level o f m RNA and protein.Results: 1.Expression quantity of Hoxa11 in mice`s endometrial stromal cells at the site of the blastocyst implantation increased with the pregnant days, as well as ?-catenin. 2.Levels of wnt4 and ?-catenin m RNA and protein were dramatically decreased in THESCs transfected successfully with lenti/Hoxa11-sh RNA compared with control ones(P<0.05).Conclusion: 1. Hoxa11 and ?-catenin positively regulates the endometrial receptivity. 2. Hoxa11 may regulate Wnt/?-catenin signaling pathway in affecting endometrial receptivity.
