Objective: Without destroying any animal physiology anatomy and giving in any packing treatment, Just inoculating staphylococcus aureus into the maxillary sinus cavity through the nasal endoscopic,to establish a new model of acute bacteria sinusitis.This model for the sake of better simulating the human body onset of acute sinusitis and promoting the further research of bacteria sinusitis.At the same time,explore the role of damaging cilia in the process of bacteria biofilm formation.Method:This was a randomized trial,eighteen New Zealand white rabbits were divided into three groups,names for A,B and C.After anesthesia,in the group A and B,every rabbit was inoculated the staphylococcus aureus in the maxillary sinus cavity through the nasal endoscopic,the amount of the bacteria respectively were 0.2 milliliter and 0.5 milliliter.The group of C,acting as control,without any operation.Following sacrifice at 7 days and 14 days after the CT of paranasal sinus,for detecting the inflammation.Then using too much anesthetic to put mices dead, nasal lavage culture was completed for detecting staphylococcus aureus. All rabbits were removed of the right-side maxillary tissue.Some tissue was disposed fixation,dehydration,embedding,section and dying, optical microscopy was used to find the histologic characteristics. Some tissue was disposed especially fluorescence stanning, confocal laser scanning microscopy was used to detect the biofilm. Some tissue was disposed fixation, scanning electron microscopy was used to observe cilia.Results: The group B,according to histopathological examination and CT scan of paranasal sinus,producing purulent sinusitis in virtually 100% of animals.The nasal lavage was cultivated on the blood AGAR palte,then the staphylococcus aureus were detected.Confocal laser sanning microscopy was used to find if they were biofilms in the paranasal sinus mucosa.Biofilm formation wsa found on the 7th and 14 th days with growths rates of 33.3%?66.7%,respectively. Scanning electron microscopy found desquamated cilia and goblet epithelium cells metaplasia,what is more,the damage of cilia in day 7 were lighter in degree than those in day 14.Optical microscopy found cilia desquamating,cells and glands hyperplasia in irregular arrangement,what is more,abundant neutrophils infiltration.As time goes,the changes in histopathological in maxillary sinus mucosa were the same as the results in scanning electron microscopy.Conclution: The rhinogenic model of acute sinusitis has been successfully established in the rabbits,which can simulate the human body onset of acute sinusitis and repeatability.At the same time,detecting the role of cilia in the formation of bacteria biofilm.The experiment proved that the demage of cilia was an important part in the process of the biofilm.We believe that it is valuable to take an consideration of recovering the function of cilia in the treatment of the bacteria biofilm in the future.
