Objective: We investigate the expression level of IL-33 in skin lesions of patients with psoriasis. And we further analysis the correlation between the IL-33 and NF-?B as well as K i-67 to explore the relationship between IL-33 and psoriasis epidermal keratinocytes proliferation. It is aimed at exploring the role of IL-33 in pathogenesis of psoriasis.Methods: We collected 28 cases skin lesion samples from patients with psoriasis and 12 cases skin tissue samples from normal contro ls. Using immunohistochemical method to detect the expression of IL-33 in Skin samples from psoriasis patients and healthy controls, and compare the difference levels of the expression of NF-?B and Ki-67 between the experimental group and control group. In addition, using the statistics to analysis the correlation of IL- 33 and NF-?B as well as IL- 33 and K i – 67 to investigate the relationship between IL- 33 and cell proliferation.Results: 1. In the 28 psoriasis paraffin specimens, IL- 33 were largely expressed in spinous layer and basal layer of epidermis in psorasis, and had a expression in inflammatory cells of dermis, but was only expressed a few in basal layer of epidermis in control group. 2. In psoriasis skin tissue samples,NF-?B were mainly expressed in epidermal basal layer, spinous layer,granular layer cell nucleus and cytoplasm,cytoplasm staining is relatively weak,but normal control group skin base layer has a small expression.The expression of IL-33(43.68%±19.50%), NF-?B(38.64%±13.96%), K i – 67(41.43%±12.49%)in psoriasis was significantly higher than that of normal control group(7.42%±4.10%,15.33%±5.71%,12.92%±5.11% respectively). 3. There were significantly positive correlations between IL-33 and NF-?B expression(r=0.583;P?0.05)or between IL-33 and K i-67 expression(r=0.587;P?0.05)in psoriasis.Conclusion: The expression of IL-33 in patients with psoriasis was higher compared to healthy controls, the expression of the NF-?B was higher than normal control group. There was a positive correlation between IL-33 and NF-?B as well as Ki-67. IL-33 may be involved in the over proliferation of epidermal keratinocytes through NF-?B and K i-67. And it may play a role in pathogenesis of psoriasis and supply a new idea for the theropy of psoriasis.Objective: We investigate the expression level and distribution of IL-33 in skin lesions of patients with Scleroderma and discuss the role in the pathogenesis of scleroderma.Methods: The expression of IL-33 and NF-? B was examined by immunohistochemical analysis in scleroderma biopsies obtained from 20 patients and 10 normal controls. And we compare the expression level between the experimental group and control group. In addition,we analysis the correlation of IL- 33 and NF-? B.Results: 1. In the skin tissue of scleroderma, IL-33 was mainly expressed in spinous laye r and basal layer of epidermis, and had a little expression in dermis. Normal control group skin spinous layer has a small amount of expression of IL-33. The positive rate of IL-33 in scleroderma was 90%, and the positive rate was only 33% of normal control group. There was a obvious difference between the expression of IL-33 in scleroderma skin tissues and the normal skin tissues. 2. In psoriasis skin tissue samples, NF-? B were mainly expressed in epidermal basal layer, spinous layer and granular layer, a part of nucleus staining is relatively weak,but normal control group skin base layer has a small expression. There were significantly positive correlations between IL-33 and NF-? B expression(r=0.774;P?0.05).Conclusion: The expression level of IL-33/NF- ? B in patients with scleroderma was obviously increased.IL-33 may be involved in the pathogenesis of scleroderma and had a certain effect for promoting skin fibrosis. IL-33 may provide new possible targets for the theropy of scleroderma.
