Objective: By selecting sand blasting zirconia, hot-etchding zirconia as experiment group, untreated zirconia and smooth surface of the cover glass as control groups, to study the biocompatibility of human gingival fibroblast(HGF) with different surface morphology of zirconia, and to verify a proper zirconia implant surface treatment for HGF adhesion and growth. In order to provide some experimental basis for the clinical application.Methods: Experiment were divided into A?B?C?D four groups: A group was sandblasting group, B group was hot-etching group, C group was control group, D group was cover glass group, which were observed by scanning electron microscopy(SEM).HGF was cultured in vitro from healthy oral gingival tissue, the cell morphology was observed by inverted microscope and identified by immunohistochemistry. HGF were used to observe the adhesion and proliferation on the surface of the material by SEM at 3h?6h?12h?24h; to measure the proliferation attached to the samples by CCK-8 at 1d?2d?3d?4d?5d?6d?7d.Results: Zirconia sandblasting, hot-etching surface with different degrees of pits wew observed by SEM, which compared with the control group were shallow and more uniform, compared with the cover glass suiface were slightly rough; HGF cultured successfully in vitro, which were adherent growth, long spindle shaped, irregular shaped, cell body refraction strong, and arranged in a radial shape, spiral shape, radial shape, also immune identification confirmed that cells derived from the mesoderm, which consistent with the appearance of fiber cell; HGF can adhesion, spreading very well on the four groups of sample surface, cell on cover glass and sand blasting surface extension was better, hot-etching surface and untreated control group was attached slower, pseudopodia fewer and shorter; HGF proliferation was in good condition, which was as same as HGF culture growth curves in vitro basically, the law of optical absorption value was cover glass group>sandblasting group> hot-etching group>control group.Conclusion: 1. HGF with zirconia has good biocompatibility; 2. sand blasting treatment is superior to hot-etching zirconia, to nontreatment zirconia, can improve HGF adhesion and growth efficiently.
