Objective: Application of rat middle cerebral artery ischemia / reperfusion model to simulate human ischemia diseases.To study the effect of remote ischemic post-conditioning on the expression of neural factors after cerebral ischemia.Methods: fifty four SD male rats were randomly divided into three groups: sham operation group(only exposed the right common carotid, external carotid artery and no middle cerebral artery occlusion and remote limb ischemia after adaptation operations), control group(using suture method to make rat cerebral artery occlusion model, 90 min after reperfusion was given and not given distal limb ischemia after adapt to the operation), experimental group(in the brain artery occlusion model 90 min underwent re perfusion give limb remote ischemic postconditioning operation). In the control group, 24 h, 72 h and 4W at different time points were divided into 3 sub groups at different time points. The experimental group was divided into 3 subgroups at different time points after 24 h, 72 h and 4W at different time points after reperfusion. Bederson nerve function score was used to score and stay in brain tissue, cerebrospinal fluid and blood samples, and the expression of MMP-9, VEGF, BDNF and GDNF were measured by Elisa.Results: 1 rats in the experimental group, 24 h and 72 h subgroup Bederson score than the control group were lower, the difference was statistically significant(P<0.05). 2, experimental groups of rats brain tissue, blood and cerebrospinal fluid were detected in VEGF and BDNF, GDNF was compared with the control in the high dosage group, MMP-9 was lower than that of the control group, with statistically significant differences(P < 0.05), and the difference in 24 h subgroups are more obvious.3 experimental groups of rats brain tissue, blood and cerebrospinal fluid were detected by western blot in VEGF and BDNF, GDNF was compared with the control in the high dosage group, MMP-9 was lower than that of the control group.Conclusion: 1, distal limb ischemic postconditioning can reduce nerve function score after cerebral ischemia in rats. 2, One of the protective mechanisms in the distal limb ischemic postconditioningthis after cerebral ischemia might be increasing VEGF, BDNF, GDNF and other factors of neuroprotective substances and reducing MMP-9 nerve damage factor.
