Objectives To evaluate efficacy of LRRK2 gene mutations on cognition function of Parkinson's disease with a Meta-Analysis.Methods There are 7 database comprehensively rearched,including Cochrane Library,MEDLINE,Embase,CNKI,WanFang Data,VIP Data and SinoMed.Prior to March 2016, the published or unpublished literatures which involve in the efficacy of LRRK2 gene mutations on the cognitive function of Parkinson's disease were included.And literatures were demanded into study according with the inclusion and exclusion criterion.All inluded clinical trials were conducted by RevMan5.3 software to evaluate the efficacy on PD patients carrying LRRK2 gene mutations(called LRRK2+ PD group) and not carrying LRRK2 gene mutations(called LRRK2- PD group) on the general cognitive function and special cognitive domains,such as language, executive ability, visualspatial function, etc.Results A total of 10 studies included in the analysis.Results show that in terms of the general cognitive function, using simple mental state examination(MMSE) as evaluation method,there has no statistically signigicant differences between LRRK2+ PD group and LRRK2- PD group.It can be think of LRRK2 gene mutation is not the cause of the general cognitive function decline.There has statistical significance between LRRK2+ PD group and LRRK2- PD group in language fluency,executive ability and visualspatial function,respectively using animal fluency test(AFT), Phonological fluency test(FAS), Stroop Word Reading, Benton judgment of line orientation test(JLOT) as the assessment methods.The result shows that LRRK2+ PD group is obviously better than in LRRK2- PD group in the language fluency, executive ability and ability of visualspatial function.Conclusion Based on the meta-analysis,carrying LRRK2 gene mutations will not affect the general cognitive function in Parkinson's disease, but may be associated with the language fluency,executive ability and visualspatial function.On the quality restrictions and publiacation bias of the current study,more studies with high quality are required to assess the results.
