Objective:Compare and analyze the gene mutations on EGFR、HER-2、PIK3CA、P53 in esophageal squamous carcinoma, analyze and discuss the genetic testing in the clinical application value of esophageal squamous carcinoma and prognosis analysis.Methods:Collected clinical data from January 2013 to December 2015 at the First Affiliated Hospital of Fujian Medical University for treatment of 112 cases of esophageal squamous carcinoma of EGFR, HER-2, PIK3 CA and P53 gene mutations, according to genetic typing divide EGFR, PIK3 CA, HER-2 and P53 into four groups, compare the differences between gender, age, location, smoking, drinking, lymph node metastasis, differentiation, and clinical stage in different gene mutation type.Results:112 cases of esophageal cancer, EGFR mutations were found in four cases(3.6%), HER-2 mutations were found in 3 cases(2.8%), PIK3 CA mutations were found in seven cases(6.3), P53 mutations were found in 66 cases(58.9%). EGFR mutations was significantly associated with lymph node metastasis(p <0.05), but not with gender, age, location, smoking, drinking, differentiation and clinical stage(p> 0.05). HER-2 mutations was significantly associated with location, lymph node metastasis, differentiation and clinical stage(p <0.05), and had a suspicious related with age(p = 0.051), but not with sex, smoking and drinking(p> 0.05). PIK3 CA mutations was significantly associated with age, smoking and clinical stage(p <0.05), but not with gender, location, drinking, lymph node metastasis and differentiation(p> 0.05). P53 mutations was significantly associated with smoking(p <0.05), but not with gender, age, location, drinking, lymph node metastasis, differentiation and clinical stage(p> 0.05). No significant correlation between EGFR, PIK3 CA, HER-2 and P53 gene mutations in esophageal cancer.(p> 0.05).Conclusion: EGFR, HER-2, PIK3 CA and P53 gene mutations had closely related to the occurrence and development in esophageal cancer. Genetic test had a great significance in early screening, diagnosis and prognosis in esophageal squamous carcinoma.EGFR mutations was significantly associated with lymph node metastasis(p <0.05), HER-2 mutations was significantly associated with location, lymph node metastasis, differentiation and clinical stage(p <0.05), PIK3 CA mutations was significantly associated with age, smoking and clinical stage(p <0.05), P53 mutations was significantly associated with smoking(p <0.05), So it had greater significance for esophageal cancer patients to test EGFR, HER-2, PIK3 CA gene, easy to find high-risk patients and to intervene in time, so we can improve the patients’ survival rate and quality of life.
