Objective The aim of this study is to explore the effect of s RAGE on RAGE mediated spiral ganglion cell apoptosis and the influence of the RAGE receptor expression in C57 mice.Methods SGCs were isolated by using a combinatorial approach of enzymatic digestion and mechanical separation from P3-5 C57 mice cochlear. After 5 days, cultured SGCs were fixed by 4?paraformaldehyde in room temperature and then immunofluorescence identification was performed by methods of S-P and the monoclonal antibody of mouse neurofilament protein(NF200). SGCs of the 5th day in vitro were randomly divided into blank control group and four groups with the same AGEs and different s RAGE as experimental groups(the concentrations of s RAGE in the medium were 0mg/L,50mg/L,100mg/L,150mg/L),and another three groups were conducted by only s RAGE used(the concentrations were 50mg/L,100mg/L,150mg/L), every groups were cultured for 24 h. Collect SGCs from different groups respectively and observe the cells of each group with transmission electron microscopy. The apoptosis rates were observed by TUNEL, m RNA expression of RAGE was detected by Real time q PCR. The statistic was deal with SPSS statistical software.ResultsThe primary culture of SGCs was successful. The quantity and activity is good enough to be examined. The cytoplasm and neurites of SGCs were dyed green with the monoclonal antibody of NF200 by immunocytochemistry Methods, and cytoplasm was deeper than neurites. SRAGE can reduce the apoptosis of SGCs mediated by RAGE, and apoptosis rates will reduce gradually when increase the concentration of s RAGE. Meanwhile RAGE m RNA expression was enhanced when s RAGE increased.Conculsion SRAGE can reduce the apoptosis of SGCs mediated by AGEs-RAGE and the expression of RAGE, this process may occur outside the SGCs,which leads to the AGEs increased before binding to the RAGE.
