Objective:In this study, we examine the expression about the lipoxin A4 receptor(LXA4-receptor) and the tumor necrosis factor-α(TNF-α) in Placenta of preeclampsia and normal pregnant women,we measured the level of lipoxin A4 and TNF-α in the blood serum of preeclampsia and normal pregnant women, to investigate the relationships between them on the preeclampsia disease, and to provide the new method of early clinical treatment for the preeclampsia disease.Methods:Select sixty pregnancy women, who delivered from December 2013 to January 2015 in the Obstetrics and Gynecologthe of the First Affiliated Hospital of Fujian Medical University and the Second Hospital of Fuqing city,were included in this study,the birth outcomes is cesarean.Study group:women with preeclampsia(n=40,mild=20,severe=20);Normal group:c(n=20).Compare their clinical feature.Use the immunohistochemical streptravidin–biotin peroxidase(SP) method compares the expression and positioning about the LXA4 receptor and TNF-α protein in placenta.Levels of LXA4 and TNF-α in the blood serum were measured by ELISA.Results:1.Each research subject’s age 、 weight 、 blood pressure and gestational weeks.(P>0.05),there are no statistical differentce between them,but the 24-hour urinary protein and CRP in the content of the preeclampsia group was significantly higher than the normal pregnant women in the control group,the difference was statistically significant.(P<0.05). 2. LXA4 receptor and TNF-α were expressed in placentas from the preeclampsia and the normal group.Immunohistochemical staining for LXA4 receptor protein was mostly expressed in villous trophoblastic cells and blood vessel endothelial cell,witch was located at the cytoplasm; Compared with mild preeclampsia and severe preeclampsia group,there were no significant difference about the expression of LXA4 receptor between them(P>0.05),the expression of LXA4 receptor in the placental tissues showed lower in the preeclampsia group than in the normal pregnant group.(P<0.05). TNF-α was mostly expressed in trophoblastic cells and blood vessel endothelial cell,partly was expressed in perivascular stroma and decidual cells,witch was located at the cytoplasm and/or epicyte.There were no statistical difference between the women with severe preeclampsia and the women with mild preeclampsia. The expression of TNF-α in the placental tissues showed higher in women with preeclampsia than in normal pregnant group.(P<0.05). 3. LXA4 receptor and TNF-α were expressed in the blood serum from the preeclampsia and the normal group.The levels of lipoxin A4 in women with mild preeclampsia higher than in normal group.(P<0.05). Compared with the severe preeclampsia group and the normal pregnant group,there were no significant difference about the expression of LXA4 between them(P>0.05). The expression of TNF-α in the blood serum showed higher in women with preeclampsia than in normal group.(P<0.05). The expression of TNF-α in the blood serum showed higher in women with severe preeclampsia than in mild preeclampsia.And the expression of TNF-α in the blood serum showed higher in women with mild preeclampsia than in normal pregnancy.(P<0.05).There were negative correlations between LXA4 and TNF-α in the blood serum of mile preeclampsia and severe preeclampsia.(MPE: r=-0.762,P<0.001,SPE: r=-0.853,P<0.001).Conclusion:1.The levels of lipoxin A4 was higher in mild preeclampsia than normal pregnancy,but there were no significant changes in severe preeclampsia group and the normal group,we suggest the reason may be attribute to the certain stress.The decreasing about the expression of LXA4 receptor in the Placenta of preeclampsia,it suggest that the defection of anti-inflammatory mediators LXA4 and LXA4 receptor would serve to aggravate the illness of preeclampsia. 2. TNF-α,as an inflammatory mediator,was higher expressed in the placental tissues and the blood serum of preeclampsia than normal pregnancy.The expressions of TNF-α in the placental tissues and the blood serum were higher in patients with severe preeclampsia than in patients with mild preeclampsia,it shows that exaggeration inflammation would serve to preeclampsia. 3.The defection of LXA4 receptor and the increase of TNF-α in the placental tissues of severe preeclampsia patients,and the correlations between LXA4 and TNF-α in the blood serum of preeclampsia is negative correlations,it hinted both the two cell factors may be interrelated in the occurrence and development of preeclampsia.
