The Impact Of Zhong Lun A Tang On MMP-3 And MMP-9 In Rats With Collagen Induced Arthritis

Objective To observe the effect of Zhonglun'e Decoction a Mongolia medicine,on the expression of Matrix metalloproteinase 3(MMP-3) and Matrix metalloproteinase 9(MMP-9) in rats with collagen induced arthritis(CIA) to to provide new strategies for targeting therapy of rheumatoid arthritis and to provide scientific evidence for Mongolian medicine to standardized treatment of rheumatoid arthritis at the same time through the impact of Mongolian medicine Zhonglun-A potions on MMP3 /MMP9 that exists in collagen-induced arthritis(CIA) rat serum or arthritis.Methods To establish reproducible collagen induced arthritis(CIA) models in rats, and set up the normal control group and Glycosides tablets group(dose of 9.45mg·kg-1), model group, Mongolian high( dose of 5.4g · kg-1),medium( dose of 2.7g · kg-1) and low dose group( dose of1.35g·kg-1).Feeding drugs for 4 weeks, HE observed synovial tissue pathologic morphology of the organization.MMP-3 and MMP-9 in the synovial fluid were measured by enzyme linked immunosorbent assay(ELISA). The distributions of MMP-3 and MMP-9 in the synovial membrane were analyzed using immunohistochemistry and Western blot.Results Compared with healthy controls, the levels of MMP-3 and MMP-9 was significantly increased in model group(P<0.01);and there was no significant difference between Zhong lun Tom high dose group with Tripterygium Glycosides(P>0.05). Positive expression of each drug group compared with the model group were reduced to varying degrees(P<0.05). The results of immunohistochemistry method showed that,the expression of MMP3?MMP9 in mode group was significantly higher than that in normal control group(P <0.01).The positive expression of MMP3?MMP9 in Glycosides tablets group and zhong lun Tom positive expression of high dose group was obviously higher than other treatment group(P <0.01).The results of Western blotting showed that the expression of MMP3?MMP-9 in the normal control group was significantly lower than that in model group and treatment group(P <0.01);the medium and low dosage of Mongolian medicine are poorer than the Tripterygium Glycosides(P<0.05).Conclusion Zhonglun-A potions has obvious improvement of CIA rat in general condition,good effect of promoting blood circulation and removing blood stasis, also it can obviously improve rat's diet and gain them weight, to a certain extent to relieve arthritis symptoms and reduce the lesion rats foot thickness,a measurable to reduce MMP-3?MMP-9 both in serum and synovial tissue. Zhonglun'e Decoction could significantly decrease the rat serum MMP-3 and MMP-9content, reduced Positive expression of synovial cells, its mechanism may be associated with it.
