Objective Nowadays the pathogenesis of henoch-schonlein purpura(HSP) has not been clear, but numerous studies have found that HSP has certain genetic susceptibility in different genetic background between different races and nationality. As genetic sequencing and genotypic technology leap development, the researches of HSP about genetics are more and more, more and more comprehensive. At present, there are some different degrees of the reseaches in different regions and nationalities. Due to the special environment of the Inner Mongolia, and the Han nationality and other nationalities are living in this region, so the study between the Han nationality and Moggol nationality has certain significance and clinical guidance which is associated with the differences of HSP. By means of researching the gene polymorphisms of Macrophage migration inhibitory factor(MIF) in the Inner Mongolia region in this article, the primary research is finding various genotypes of MIF(CC, CG, GG)between the Han nationality and Moggol nationality, to find that whether MIF-173 gene loci is associated with HSP of the Han nationality and Moggol nationality, and MIF-173 gene loci whether have any differences between the Han nationality and Moggol nationality or not.Methods The cases of this research were found in Inner Mongolia medical university affiliated hospital and Inner Mongolia International Mongolian hospital since December 2012- December 2013. There is each 25 cases of the Han nationality and Moggol nationality with HSP. Among them, the Han nationality boy 16 cases, girl 9 cases, Moggol nationality boy 11 cases, girl 14 cases. We found control groups in Inner Mongolia medical university affiliated hospital and Inner Mongolia International Mongolian hospital, 30 cases of the Han nationality and 25 cases of Mongol nationality. Among them, 18 cases of the Han nationality boy, girl 12 cases, 13 cases of Moggol nationality boy, 12 of Moggol nationality girl, and age distributed from 1 to 14 years old. The experiment divided into four groups: Moggol nationality with HSP group and the group of the Han nationality with HSP, Moggol nationality control group and Moggol nationality control group. Each sample was drawed from children's venous blood2 ml on an empty stomach, and put in EDTA anticoagulant tubes in- 80 ? refrigeratorwaiting for inspection. We could use DNA Extraction Kit to extract all DNA and used the method of polymerase chain reaction- restriction fragment length polymorphism(PCR-RFLP) to detect the polymorphism of MIF 173 gene locus in the two nationalities. At last,we used appropriate statistical methods to calculate each genotype's frequency, and then compared them using SPSS17.0 statistical software. The experiment compared the polymorphism of MIF 173 gene locus in these groups: Moggol nationality with HSP group and the group of the Han nationality with HSP, Moggol nationality with HSP group and Moggol nationality control group, the Han nationality with HSP and the Han nationality control group. As ?=0.05, P?0.05 for the standard, if P?0.05, the difference is significant statistically, if not, the difference is not significant statistically. According to Hardy weinberg theorem we could calculate MIF 173 gene locus(CC, CG, GG) each gene's frequency.Results MIF-173 gene loci could detect the three genotypes, namely, GG, CG and CC type. Compared to the group of the Han nationality with HSP and the Han nationality control group, there is mostly MIF-173 CC gene loci; the two groups of different genotypes compared with each other, ?2 =10.459, P=0.005, the difference is significant statistically.Compared to Moggol nationality with HSP group and Moggol nationality control group, there is mostly MIF-173 CC gene loci; the two groups of different genotypes compared with each other, ?2 =6.607, P=0.037, the difference is significant statistically, too. However, compared to Moggol nationality with HSP group and the Han nationality with HSP group, ?2 =0.099,P=0.952, the difference is not significant statistically.Conclusion MIF is likely to play a important role in the occurrence and development of HSP, and MIF-173 C allele gene has a close relationship with the Han nationality and Moggol nationality about HSP; MIF-173 C allele gene is a susceptibility gene. There is no difference between the Han nationality and Moggol nationality about HSP in MIF-173 gene loci polymorphism. It is suggesting that MIF-173 gene loci polymorphism is not the reason which can lead to different clinical feature between the Han nationality and Moggol nationality about HSP.
