Objective A rat learning and memory impairment model was established by long-term and heavy drinking with brick tea. Exceptly, the index of the bone metabolism in the serum was also detected. The purpose of the study was to investigate the mechanism of its occurrence and provide a theoretical basis for the preventation and treatment about the F-Al combined toxicosis caused by drinking brick tea.Methods Wistar rats were 5-week, 7 groups were divided randomly, 10 rats in each group,male and female were both half. Normal control group(drinking water), fluorine group(flourine-50mg/Lsodium fluoride solution), aluminum group(aluminum 163 mg/L aqueous solution of aluminum chloride), fluorine and aluminum group(fluorine-50mg/L,aluminum-163mg/L, Mixture),high-dose fluoride-aluminum brick tea group(flourine-50mg/L,aluminum-163mg/L, brick tea water),low-dose fluoride-aluminum brick tea group(flourine-10mg/L, aluminum-33mg/L, brick tea water),Low fluorine and low aluminum brick tea group(flourine-2mg/L,aluminum-1.5mg/L brick tea water).General state regularly observed in rats. Morris water maze was used to test the ability about learning and memory in rats at the end of 12 months. MDA, T-SOD, GSH-PX, GST of Brain was detected,Immunohistochemistry and Real Time-PCR method were used to detect the protein and m RNA expression of bcl-2/bax in hippocampus neural cells. In addition, Elisa was used to detect PTH, OT, CT of the serum, ALP, Ca, and P were also detected.Results Learning and memory impairment : Fluorine group, aluminum group, fluorine and aluminum group and high-dose fluoride-aluminum brick tea group can cause dysfunction of leaning and memory(P<0.01), Aluminum group is the most serious(P<0.01), The antioxidant levels of fluorine group, aluminum group, fluorine and aluminum group is lower than normal control group(P<0.01), aluminum group is the lowest(P<0.01); The antioxidant levels of fluoride brick tea group is also reduced(P<0.05), but higher than the fluorine and aluminum group(P<0.01), The lipid peroxidation products of fluorine group and aluminum group is increased(P< 0.01). Fluorine group and fluorine and aluminum group can Lower the protein and m RNA expression of bcl-2/bax, fluoride brick tea group also can lower the protein and m RNA expression of bcl-2/bax(P<0.01). The vitality of T-SOD(P<0.01) and GSH-PX(P<0.05) in low fluorine and low aluminum brick tea group is higher than normal control group, and the protein expression of bcl-2/bax is still higher than normal control group(P<0.05).Tooth and serology: The tooth of normal control group is brown, smooth and shiny;different degrees of dental fluorosis was found in fluorine group, fluorine and aluminum group and high-dose fluoride-aluminum brick tea group; no dental fluorosis was found in aluminum group; only one mild dental fluorosis was found in low fluorine and low aluminum brick tea group. PTH, OT in fluorine group, fluorine and aluminum group and high-dose fluoride-aluminum brick tea group were significantly higher(P<0.01) than the control group,and the highest is fluorine group; PTH, OT, ALP in aluminum group is lower than the control group(P<0.01); Ca in fluorine and aluminum group and brick tea group is lower than the control group(P<0.01); CT in fluorine group, fluorine and aluminum group and high-dose fluoride-aluminum brick tea group is lower than the control group(P<0.01), fluorine and aluminum group is the highest, but the aluminum group is increased.Conclusions 1.Morris water maze test have confirmed the dysfunction of leaning and memory caused by drinking brick tea is F-Al combined toxicosis. 2.The dysfunction of leaning and memory caused by drinking fluoride brick tea with a long time in rat is correlate to the anti-apoptotic ability of hippocampal neurons. 3.Oxidative stress is also involved in the mechanism of learning and memory injury. 4.Low-dose fluoride-aluminum brick tea group can cause learning and memory impairment. 5.Low fluorine and low aluminum brick tea has good ability of antioxidant and anti-apoptotic, Therefore we can advocate to drink low fluorine and low aluminum brick tea. 6.The injury induced by fluoride is more obvious, the antagonism maybe exist between fluoride and aluminum, the bone injury was indirectly reacted by serum biochemical indicators, Relatively speaking, OT, PTH, CT and ALP is more sensitive.
