Objective To observe the 3D Spheroid Cultured Mesenchymal Stem Cells transplantation on cerebral ischemia-reperfusion injury in rat tissue tumor necrosis factor(TNF-?), interleukin-6(IL-6) and interleukin-10(IL-10). Discussing the possible nerve protective mechanisms of stem cells in rats with Mesenchymal Stem Cells after ischemia.To provides a theoretical basis for the clinical application of Mesenchymal Stem Cells transplantation in the treatment of cerebral ischemic injury.Methods The experimental animals were randomly divided into sham group,control group, MSCs treated group. Suture method is used to establish the rat middle cerebral artery occlusion(MCAO) model. Unplugging the fishing line for reperfusion in 2h after ischemia, using neurological score to select model, observe the transplantation distribution of labeled MSCs in hippocampal CA1 region by Fluorescence microscopy.The rats were decapitated after the intervention1 day, 3 day and 7 day. Using the Nissl's staining to observe the neurons changes in ischemic area. Using the method of enzyme-linked immunosorbent assay(ELISA) and RT-PCR to detect the variation of rat brain tissue TNF-??IL-6 and IL-10 expression.Results MSCs labeled express in hippocampal CA1 region; compared with the control group, Treatment group can improve the motor function of rat significantly on the seventh day(F=29.996, t=2.236, n=6, p<0.05), Nissl's staining show that neuronal populations increase and the form is complete relatively. The content of TNF-? and IL-6in brain tissue of MSCs group were significantly lower by ELISA(F=112.525~362.381,t=8.053~22.268,P<0.01), while the brain tissue IL-10 expression was significantly increased(F=289.593~976.587, t=10.740~28.449, P<0.01).RT-PCR results: The TNF-? and IL-6 m RNA expression of MSC group rats was significantly lower than control group(F=116.502 ~116.502, t = 466.271~23.168, P <0.01), and brain tissue IL-10 m RNA expression higher than the control group(F = 80.492~ 80.492, t = 582.995~16.912, P < 0.01).Conclusion MSCs has neuroprotective effects and its mechanism may be associated with reducing brain tissue TNF-?, IL-6 levels, increased IL-10 content.
