| Objective:During the process of liver transplantation,the donor liver mostly through a low temperature storage period, during this period the liver cells will be damaged, and the damage can increase over time, the damage will directly affect thequality of the donor liver, how to reduce this injury is very important during liver transplant. Danshen in the University of Wisconsin solution can provide an good effect to the donor liver, it is a question to explore the concentration difference of the effect ofthe University of Wisconsin solution。Methods:(1)Experimental materials and grouping: 16 male SD rats were used, high fat diet for 3months, all moderate fatty liver model,divided into model control group(the University of Wisconsin solution cold preservation group), 200 mg, 400 mg / L, 600 mg / L Danshen in the University of Wisconsin solutiongroup according to the random number table method,four rats in each group.(2) Experimental methods: preparation rat donor liver warm ischemia model, donor liver were placed in different concentration of Danshen inthe University of Wisconsin solution,The microdialysis catheter was placed in the liver of microdialysis sampling, interval of 20 minutes to take a sample, continued to collect12 h, the glucose, lactic acid, pyruvic acid, and glycerolfrom the microdialysis samples were measured.At the same time,comparate the observation for liver at different times of the pathological morphological changes.Results:The model control group and the experimental group were all analysised, without drop out.(1) The levels of glucose and glycerol of the test groups and model control group continue to rise during the cold preservation process。The test groups up to a lower level than the model control group, and the 400mg/L group have the lowest level.(2)The inflammatory cell infiltration and necrosis cells of model control group are less than the test groups,and the 400 mg/L group liver cell necrosis lightest.Conclusion:(1) Danshen in the University of Wisconsin solutionhas protective effect on liver transplantation, and the400 mg / L group is best.(2)We can evaluate the liver according to the level of lactate, lactate / pyruvate ratio and glycerol by the use of microdialysis. |
