Objective:To explore the curable effect of PND on mice with immune mediated apalstic anemia and its regulation of T lymphocyte cytokines and transcription factors including ROR-?t,FoxP3,we may approach the effect of PND on immune regulation function.Methods:60 SPF level BALB/c mice were selected for this study and randomly divided into six groups,namely,normal control group,the irradiation group,the aplastic anemia model control group,PND low and high dose treatment groups,and the cyclosporine group.The aplastic anemia model mice were built with immune-mediated method.Observe the peripheral blood leukocytes and platelets of tail vein blood of mice on the 7th day and the 14th day;Killing the mice on the 14th day,take bone marrow biopsy of both sides of mouse femurs and observe the bone marrow smears by the microscope.Detecting cytokine of mouse peripheral serum by CBA.Extracting the spleen protein of the mice,observed the transcription factors including ROR-yt and FoxP3 of each group of mouse spleen protein by Western blot experiment.Results:1.PND could increase the peripheral blood cells of mice with immune mediated apalstic anemia.The peripheral blood leukocytes and platelets of PND treatment group were higher than the aplastic anemia model control group.2.PND could promote the recovery of hematopoietic function of mice with immune mediated apalstic anemia.By the comparison of the biopsys,the marrow tissue sections of mice with immune mediated apalstic anemia appeared light staining of the hematopoietic tissue,extremly decrease of bone marrow hyperplasia and increase of adipocytes.With the dose increasing of PND,the hematopoietic tissue was gradually repaired,the hematopoietic cells were gradually increased and the adipocytes were significantly decreased.3.PND could regulate the cytokine levels in T lymphocytes of mice with immune mediated apalstic anemia.Compared with the aplastic anemia model control group,PND could reduce IFN-??TNF-? secretion and increase IL-4?IL-10 secretion.4.PND could decrease the expression of ROR-yt and increase the expression of FoxP3,that is,promote the recovery of the imbalance of Th17/Treg.Conclusion:PND can improve the peripheral blood and bone marrow of mice with immune mediated apalstic anemia.PND could promote the recovery of the imbalance of Thl/Th2 and Th17/Treg,.and regulate the T-lymphocyte Immune function.
