| Objective:To investigate the expression of aquaporin 1,5 in the Eustachian tube mucosa of a genuine pig model of otitis media with effusion(OME) caused by surgical obstruction of the Eustachian tube.Methods:Thirty-five genuine pigs underwent unilateral Eustachian tube obstruction. We performed otoscopic evaluation after the surgery and divided these genuine pigs into four groups- 3d,7d,14d,28d. The genuine pigs were sacrificed after otoscopic evaluation, and their temporal bone were harvested. Hematoxylin-eosin staining was done to recognize the morphological changes of the Eustachian tube mucosa. Immunohistochemistry and Western-blot were done to find out the locate and change of AQP 1,5.Result:Twenty-seven genuine pigs were successfully operated and finally the number of the genuine pigs qualified in group 3d,7d,14d,28d was 6,5,4,3, respectively. The Eustachian tube mucosa showed substantial histologic changes since the onset of OME. Partial of the cilia epithelium underwent defect and necrosis. Then recovered along with the OME. AQP 1 was located in fibroblasts and vascular endothelial cells. AQP 5 was located in the apical of ciliated cells and glands. The level of AQP 1,5 expression decreased significantly since the onset of OME and slowly increased since day 14.Conclusion:1. AQP 1 and AQP 5 are found in genuine pig Eustachian tube musoca. AQP 1 was located in fibroblasts and vascular endothelial cells and AQP 5 was located in the apical of ciliated cells and glands.2. The expression level of AQP 1 and AQP 5 experienced a change that declines first but inclines afterward. We can find out the connection between the expression level of AQP 1 and AQP 5 and the extent of damage of the Eustachian tube mucosa.3. AQP 1 and AQP 5 in the Eustachian tube mucosa maintain the integrity of Eustachian tube mucosa and the function of mucociliary system, play a significant role in the onset and outcome of OME. |
