Background Bone marrow mesenchymal stem cells (BMSCs) have shown promising prospects in promoting tendon-bone healing after anterior cruciate ligament reconstruction. It has been demonstrated that there are mesenchymal stem cells existing in the anterior cruciate ligament based on the latest research. Moreover, these cells have the similar characteristic of proliferation and multi-differentiation with BMSCs. However, there are still no clear reports on the comparison of biological characteristics of these two stem cells.Objective To compare the biological characteristics of Bone marrow mesenchymal stem cells (BMSCs) and Anterior cruciate ligament derived mesenchymal stem cells (ACLMSCs) from rats in vitro.Methods Ten male SPF-level rats, weighing 200-220 grams, were prepared. Anterior cruciate ligaments and bone marrows were obtained from rats to culture ACLMSCs and BMSCs respectively under sterile condition. These cells were observed, olated, passaged purified and identified in vitro. Meanwhile, the surface marker, growth kinetics and mRNA levels of differentiation related genes (ALP, Bglap, Runx2, Bmp2, Spp1, Col2?1, Acan, Sox9, PPAR?2 and c/EBP?) were also determined.Results and conclusion Morphology was similar between BMSCs and ACLMSCs under an optical microscope. ACLMSCs and BMSCs at passage 3 highly expressed CD29, CD90 and were negative for CD45,CD34. The growth kinetics and cell clone analysis indicated that the proliferation rate of ACLMSCs was significantly faster than that of BMSCs. Both of them were induced to osteoblasts, adipocytes and chondrocytes. Differentiation related gene detection shown that ACLMSCs expressed higher levels of chondrogenic markers (Col2al, Acan and Sox9) (p=0.0003, p=0.0020, p=0.0064).These findings suggest that the proliferative potential of ACLMSCs is significantly greater than that of BMSCs,and the former is apt to the chondrogenitic differentiation. ACLMSCs are promising cells to promote tendon-bone healing.
