Objective To investigate the effect of Centella Asiatica on expression of Connective Tissue Growth Factor(CTGF) and Fibronectin(FN) in NRK52E cells stimulated by high glucose,and further explore the mechanism of centella asiatica on inhibiting renal tubular interstitial fibrosis and treatment of diabetic nephropathy.Methods The rat renal tubular epithelial cell line NRK52E were cultured and divided into six groups:1)Normal control group;2)high glucose group;3) high glucose group treated with high dose of centella asiatica; 4) high glucose group treated with middle dose of centella asiatica; 5) high glucose group treated with low dose of centella asiatica; 6) high glucose group treated with captopril.,Cells of each group were collected after cultured 48h and then the following experiments were carried out.1)Western blot was used to detect the expression of CTGF and FN protein; 2) Real Time PCR was used to detect the expression of CTGF and FN mRNA.Results 1)Compared with the normal control group, the expression of CTGF and FN protein of high glucose group were up-regulated obviously(P<0.05);the expression of CTGF and FN protein of different dose of centella asiatica groups and captopril group down-regulated markedly when Compared with the high glucose group(P< 0.05),the CTGF expression level of treatment groups were:middle dose of centella asiatica group<high dose of centella asiatica group< captopril group<low dose of centella asiatica group,while the FN expression level of treatment groups were:captopril group <high dose of centella asiatica group< middle dose of centella asiatica group<low dose of centella asiatica group.2) Compared with the normal control group, the expression of CTGF and FN mRNA of high glucose group were up-regulated obviously(2-??Ct>2); compared with the high glucose group,the expression of CTGF and FN mRNA of different ose of centella asiatica groups and captopril group down-regulated markedly (2-??Ct<0.5);the result of pairwise comparison showed, the CTGF mRNA expression between treatment groups had not got obvious difference, while the FN mRNA expression of captopril group was significantly lower than that of middle dose of centella asiatica group (2-??Ct<0.5).Conclusion 1)High glucose can induce the increase of CTGF and FN mRNA and protein expression in renal tubular epithelial cells, which may be one of the important mechanisms of diabetic nephropathy and renal interstitial fibrosis; 2) Centella asiatica can inhibit the increase of CTGF, FN mRNA and protein expression of renal tubular epithelial cells induced by high glucose cultured in vitro;3inhibition of (3), the mechanism of prevention of interstitial fibrosis and treatment of diabetic nephropathy of Centella asiatica may be related to inhibition of CTGF, FN expression, which will regulate the overproduction and deposition of extracellular matrix.
