Objective: To study the effect of fenugreek total flavonoids of behavioral despair depression model mice,reserpine injection induced mice model of depression and depression rats model with chronic unpredictable stimulation.To determine the number of independent activities,suspended tail activity time and swimming immobility time of behavioral despair and depression model.Detection of reserpine injection induced depression model mice eyelids droop degree,number of circles,body temperature and content of 5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain homogenate.Determination of depression rats model with chronic unpredictable stimulation body weight increase,sugar water consumption amount,the swimming immobility time and content of5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain homogenate,levels of adrenocorticotropic hormone?ACTH?,cortisol?CORT?,and Interleukin-6?IL-6?.The weight of thymus and spleen and the organ index were calculated.The pathological changes of hypothalamus and pituitary gland,thymus,spleen and adrenal gland were observed under light microscope.To reveal the mechanism and characteristics of fenugreek total flavonoids in the treatment of depression.Methods: Behavioral despair and depression mice model.Administration of fluoxetine hydrochloride solution(10mg·kg-1),high,middle and low dose of Fenugreek total flavonoid(400mg·kg-1?200mg·kg-1?100mg·kg-1)were administered to the drug group every day,the saline group was given the same volume of saline solution,nine point to the medicine every day,continue 9d.At 7d,after 1 hours of administration,determinate the number of locomotor activity of mice in 5 minutes.At8 d,after 1 hours of administration,determination of suspended tail activity time in4 min.At 9d,after1 hours of administration,the swimming immobility time wasmeasured in 6 minutes after 5 minutes.Reserpine injection induced mice model of depression.Administration of fluoxetine hydrochloride solution(10mg·kg-1),high,middle and low dose of Fenugreek total flavonoid(400mg·kg-1?200mg·kg-1?100mg·kg-1)were administered to the drug group every day,the saline group was given the same volume of saline solution,nine point to the medicine every day,continue 7d.After 1 hours of the last administration,intraperitoneal injection of reserpine?4mg/kg?,to observe the eyelids droop and the number of out of the mice in 15 s.After the 4h,with a thermometer to measure the body temperature of mice.The mice were killed,took out the brain tissues,determination the content of 5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in mice brain homogenate.Depression rats model with chronic unpredictable stimulation.At the beginning of the modeling,the experimental animals were administered with fluoxetine hydrochloride solution(10mg·kg-1),high,middle and low dose of Fenugreek total flavonoid(400mg·kg-1?200mg·kg-1?100mg·kg-1)were administered to the drug group every day,the saline group was given the same volume of saline solution,nine point to the medicine every day,continue 21 d.Determination of sugar consumption in nineteenth days.At 20 d,after 1 hours of administration,determination the swimming immobility time.After 1 hours of the last administration,determination the wet weight of rat thymus and spleen and calculate the organ index of thymus and spleen.Determination the content of 5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain homogenate and the levels of adrenocorticotropic hormone?ACTH?,cortisol?CORT?,and Interleukin-6?IL-6?in serum.The pathological condition of the corresponding viscera was observed under light microscope.Results: Behavioral despair and depression mice model.Compared with the treatment group,the swimming immobility time of the mice was significantly increased,the number of independent activities and the time of suspension tail activity decreased significantly in the normal saline group.The fenugreek total flavonoids could significantly decrease swimming time and increase the number of independent activities and tail suspension.Reserpine injection induced mice model of depression.Compared with the normal saline group,the exercise capacity decreased,the eyelids droop and body temperature decreased,and the contents of 5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain tissue decreased obviously in model group.The fenugreek total flavonoids of the gourd could improve the eyelid droop,and increase the body temperature and the contents of 5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain tissues.Depression rats model with chronic unpredictable stimulation.Compared with the normal saline group,the sport ability decreased significantly,for sugar water consumption and Viscera index decreased significantly,and the contents of5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain tissue reduced in the model group.Conclusion: Fenugreek total flavonoid large,medium and small dose group could improved locomotor activity,activity time swimming and tail suspension immobility time with behavioral despair depression model mice.Fenugreek total flavonoid could improved activity,reduced the degree of drooping eyelids,reduced swimming immobility time,raise body temperature and improve the levels of5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain tissue.For chronic unpredictability of depression in rats could significantly improved content of 5-hydroxytryptamine?5-HT?,dopamine?DA?and norepinephrine?NE?in brain homogenate and levels of adrenocorticotropic hormone?ACTH?,cortisol?CORT?,and Interleukin-6?IL-6?in serum,could significantly improved immune function,and lesions of various organs and tissues have improved greatly effect.
