Objective:To study the change rule of main components in the process of processing and decocting,and explore the efficacy before and after processing,try to reveal the processing mechanism of Semen Cassia.Methods:Established a HPLC method of simultaneously determining the content and decocting rates of 3 naphthopyrone glycosides and 3 anthraquinones in Semen Cassia seeds before and after processing by,with Dionex Acclaim C18 column was used,acetonitrile(A)-THF-1 % aceticacid(0~30min:16 %~18 % A,2% B,82 %~80 % C;30~35 min:18 %~45 % A,2% B,80 %~53 % C;35~55 min:45 %~70 % A,2% B,53 %~28 % C)was used as amobile phase,with flow rate of 1 m L·min-1,column temperature at 30 ℃ and detection wave length at 278 nm.Drench the rats raw and processed Cassia decoction,components groups and monomers with ink,observe and record the first defecation time and dung grain number,study the effect of purging efficacy of Cassia seeds processing.Use the Qxford cup diffusion method to study the antibacterial essicacy of raw and processed decoction component groups and monomers.Established a HPLC method to determin the ACE inhibitory efficacy of raw and processed decoction component groups and monomers in vitro.Results:The content of water soluble extract of Semen Cassiae increased 1.7% after processed,and raised13% after mashed,respectively.The content of 3 naphthopyrone glycosides in decoction were dropped after processed,but 3 anthraquinones were increased.The above results were similar to the samples after mashed and unprocessed,but opposite to that after mashed and processed.The decocting rate of all compounds were dropped in the samples after processed without mashed,but 3 naphthopyrone glycosides went up and 3 anthraquinones dropped in the samples after processed with mashed.The raw and processed Cassia seeds decoction exhibits significantly purging efficacy,and the processed Cassia is a bit strong,but no significant difference in statistics.Cassiaside(naphthalene and pyrone glucosides),obtusifolin-2-O-β-glucoside(anthraquinone),anrantio –obtusin(anthraquinones),raw glycosides and raw coarse both have purging efficacy,and the raw coarse is stronger.Antibacterial activity study showed that,Semen Cassia seeds decoction,methanol extracts have inhibitory efficacy on staphylococcus aureus,but not for escherichia coli,bacillus subtilis and pseudomonas aeruginosa.Afte processing,the inhibitory efficacy of decoction on staphylococcus aureus is weaker,but there is no difference between methanol extracts.The antibacterial activity of 10 aglycones and 5 glycosides shows that,5 aglycones have inhibitory efficacy on staphylococcus aureus,but 5 glycosides not.The raw and processed Cassia seeds both have inhibitory efficacy on ACE activity,and no significant difference.The most selected components in Cassia seeds have inhibitory efficacy on ACE activity.Anrantio –obtusin,chryso-obtusin,2-hydroxyemodin-1-methl-ether,Obtusifolin-2-glucoside,Physcion-8-β-gentiobioside,physcion-8-O-β-D-glucopyranoside,rubrofusarintriglucoside inhibits ACE activity is more than 30 %.Conclusion:It both had a great impact on the content of active compounds by mashed and processed.And in the processing and decocting procedure,glycosidic enzymes contained in Semen Cassiae were involved in promoting the transformation of the major active ingredients.The efficacy of Semen Cassiae is closely related to its composition.Naphthalene and pyrone glycosides and their aglycones,anthraquinones glycosides and their both have purging,antibacterial and calming Ganyang efficacy.Because the purging and antibacterial efficacy of aglycones are stronger than glycosides,after processing,partial glycosides transfer to aglycones,enhanced the purging efficacy.Because the decocting rate of activity components dropped,the antibacterial efficacy became weaker.
