At present, it shows the importance of type I collagen sponge in the aspect of clinical application. For many kinds of severe burn, especially after the dermal tissue transplant operation, it has brought great impact to people’s daily life. It is necessary that to development of type I collagen sponge which has a great significance. In this article, it involves that with the irradiation crosslinking to make type I collagen gel. And then, it will be the sponge after the freeze-drying. In this way, it abandoned the method which using the small molecule crosslinking chemical agent. After that it would be waste of much water to douche the sponge. In this article, we used the method of efficient and clean irradiation crosslinking, it has affected on the material microstructure. Except that we discuss the biological histocompatibility though the animal experiment.1. Using commercially available fresh bovine tendon to do the part of type I collagen extraction work, the dosage of pepsin and the substrate concentration have affection on the extraction efficiency. Through control the extraction temperature and pH of solution conditions, the best condition of the extraction was obtained by experiments of raw materials for testing. At last we proved that it is type I collagen.2. By controlling the different conditions of type I collagen solids as well as the irradiation dose and dose rate, after irradiation, the type I collagen gel was freeze-drying. Type I collagen sponge was tested on the material detection targets and pore size distribution, determine suitable for practical application.3. To verify the radiation cross-linked materials can be degraded in vivo through animal experiments. After that, the H&E stain, Masson stain and Chinese ink perfusion experiment showed the excepted performance in the organization biocompatible material. Evans Blue perfusion experiments proved that the material can be vascularized. The relationship between each other could be confirmed and supported each other. There is a good judgment to the organization biocompatible material.
