The Extraction And Enrichment Of Volatile Oil From Bupleurum Chinese

Objective: The composition,extraction process and enrichment with macroporous resin of volatile oil from Bupleurum chinese were systematically studied in this paper,and the best process parameters have been optimized. To innovate process on the basis of steam distillation. The problem of difficulty in oil-water separation and low extraction of volatile oil in traditional Chinese are effectively resolved in industrial production. It is aimed at providing a new idea about enriching volatile oil from Bupleurum chinese and even traditional Chinese.Methods: Firstly,the main component and relative content of volatile oil from Bupleurum Chinese were determined by GC-MS. Using hexanal, heptanal,(e)-2-octena and trans-2,4-decadienal as standard sample, a HPLC method for determination the content was established. By using the content of the four aldehydes as standard, the best extraction process of volatile oil from Bupleurum chinese was determined by single-factor test and orthogonal experiment. Based on static adsorption and desorption experiments, suitability type of macroporous resin was filtered. In addition, using single-factor test and orthogonal, the study made some research through dynamic adsorption and desorption,including flow rate of solution which contains volatile oil from Bupleurum Chinese,p H,diameter height ratio of resin and the best sampling. Using single-factor test to study the consumption of 95% ethanol for eluting. Finally,high content of volatile oil from Bupleurum Chinese was made by recycling ethanol with rotary evaporation.Results: Low molecular weight aldehydes are the main elements of volatile oil from Bupleurum Chinese as well as the main efficacy component. The optimum process of extracting volatile oil from Bupleurum Chinese: 8 times amount of water,collecting the distilled liquid of equal to 3 times amount of medicinal Herbs. The optimum process of enriching volatile oil from Bupleurum Chinese: Using 5 m L HPD-100 macroporous resin,700 ml sample solution(46.67g·m L-1), diameter height ratio of resin 1:7, sample flow rate 1.5 m L·min-1, dosage of 95% ethanol 16 m L diversion 79.71%.Conclusion: The process of extraction and enrichment of volatile oil from Bupleurum Chinese are simple, stable and highly repetitive, which can be used in mading high content of volatile oil from Bupleurum Chinese.
