Objective: To establish cisplatin-resistant gastic cancer SGC7901/D DP by concentration gradient method and explain multi-resistan corre lation between DCLK1 and gastric cancer SGC7901/DDP cellsMethods:1. cisplatin-resistant gastic cancer SGC7901/DDP was esta blished by concentration gradient method.Then we observed the m orphological changes of SGC7901/DDP through an inverted microsc ope,draw the cell growth curve and calculate the population doubli ng time of SGC7901/DDP by cell counting;The resistance index an d cross-resistance characteristics of SGC7901/DDP was detected by MTT assay,The drug resistance associate genes and proteins express of SGC7901/DDP and SGC7901 were detected by Real time Qua ntitative PCR and Western Blot; 2. SGC7901/DDP-DCLK1-sh RNA was established by RNA interference,then changes of the resistanc e index?the drug resistance associate genes and proteins express we re detected by MTT assay?Real time Quantitative PCR and Wester n Blot.Result:1.We established the drug-resistance gasric cancer cell SGC7901/DDP successfully. Compared with SGC7901, under inverted microscope gasric cancer cell SGC7901, SGC7901/DDP cells wereRound or ellipsoidal shape,showed Colony,irregular nuclea and gaint cells,the proliferation rate of SGC7901/DDP slowed down.The resi stance index of SGC7901/DDP to DDP, taxinol?5-FU were 26.495?11.9081?11.358 respectively.2.The genes and proteins of DCLK-1 and MRP1 of SGC7901/DDP cells expressed higher than those of SGC7901 cells(P<0.05).3.We established SGC7901/DDP-DCLK1-sh RNAcells.Compared with SGC7901/DDP-control-sh RNA, The genes and proteins of DCLK-1 and MRP1 of SGC7901/DDP cells expressed lower obviosily(P<0.05)4.The resistance index of SGC7901/DDP-control-sh RNA to DDP, tax inol?5-FU became small than those of SGC7901/DDP-control-sh RN A(p<0.05).Conclusion:1. Cisplatin-resistant gastic cancer SGC7901/DDP was e stablished by concentration gradient method. 2. DCLK-1 and MRP1 both expressed higherly in SGC7901/DDP, MRP1 was a positive correlation with DCLK-1. 3.Downgrading DCLK-1 expression led to the drug resistance of ga stric cancer cell decrease,which may be related to MRP1 low expre ssion.
