Objective:This study aimed to investigate the expression of Y-box binding protein-1( YB-1) in pancreatic carcinoma tissue and its relationship with clinicopathological features and prognosis.Methods:Collecting 60 cases of pancreatic carcinoma specimens with pathological diagnosis and complete follow-up data,and Collecting 15 cases of fresh tissue of pancreatic carcinoma(well-differentiated,moderately differentiated and poorly differentiated tissues each have 5 cases), then respectively using immunohistochemistry, in situ hybridization and Werstern blot for detecting the expression YB-1 protein and mRNA in pancreatic carcinoma tissues and adjacent normal tissues. Collecting pancreatic carcinoma patients with data of clinic-pathological features, and using statistical methods for analyzing the association of the expression of YB-1and patients' clinic-pathological data(age, gender, tumor size, tumor location, tumor differentiation, TNM stage, lymph node metastasis and survival time).Results: 1.Immunohistochemistry showed the positive expression rate(39/60,65.00%) of YB-1 protein in cancer tissue was significantly higher than that of the paired adjacent normal tissues(7/60,11.67%)(x~2=36.099, P < 0.001); there was a linear relationship between differentiation of pancreatic cancer and the expression degrees of YB-1 protein, and the expression intensity of YB-1 protein enhanced when the degree of differentiation decreased. 2. In situ hybridization showed similar results:the positive expression rate(28/60,46.67%) of YB-1 mRNA in cancer tissue was significantly higher than that of the paired statistically significant(P=0.045<0.5).Conclusions: The expression of YB-1 was negatively associated with the differentiation of pancreatic carcinoma, and the clinical TNM stage and lymph node metastasis was positively related to the prognosis, the median survival time of patients with negative expression of YB-1 protein is under that of positive expression, suggesting that YB-1 maybe a molecular marker for evaluating survival of pancreatic carcinoma.
