Currently, depression has been listed as one of the common diseases in the world. According to the latest statistics from the WHO, the number of people with depression in the world is up to 0.35 billion now. Depression is different from the usual mood swings, and the symptoms are very complex. Moderate or severe depression patients in the study, life and work have shown a poor state, they have often suicidal thoughts. The suicide population is estimated to be around 1 million every year. There are many disadvantages in the present antidepressant drugs, such as poor tolerance, slow onset of action, and more adverse reactions, which increases the risk of suicide. Therefore, it is a great theoretical value and practical significance to search for antidepressant drugs with high efficacy, fast acting time, good tolerance and high safety.In the paper,we designed and synthesized a series of novel multi flumorph compoundswith trifluoromethyl benzene acetone as the starting materialon on the basis of the preliminary work of our research group.The mice forced swimming pharmacological experiments were used to evaluatethe antidepressant activity of target compounds. The contents of this paper are as follows:1?Designed and synthesised 8 novel trifluoromethyl morpholine hydrochloride compound with trifluoromethyl benzene acetone as the starting materialon.The target compound structure had been characterized by 1H NMR, IR and MS spectra.2?The best reduction and acidified conditions:the reducing reaction temperature was 160? and the reflux reaction time was 12 h.3.To evaluate all synthesized compounds antidepressant activity by the forced swimming test. The structure of target compounds were identified by 1H NMR, IR and MS. The immobility time of compound Vb,Vf were 110.1 ± 27.5s,107.1 ± 29.4s, The immobility time of blank group was150.4 ± 38.5s, The immobility time of compound Vb,Vf were shortened significantly(P<0.05). The synthetic process was simple and easy to control. The test results revealed that Compound Vb and Vf have shown someantidepressant activities.
