Objective : To preparation functional health food of puerarin with hypoglycemic,lipid-lowering effect, orthogonal experiment design method were used to research puerarin flavonoids and its polysaccharide extraction process conditions. Used puerarin extract as mainly composition, optimizing prescription of puerarin chewable tablets, and the quality evaluation were analysised. Effect of puerarin chewing tablets on type 2 diabetic rats model of blood glucose, blood lipid and antioxidation were discussed.Mehods:Based on single factor experiment, effect of three factors temperature, time and solvent water on extraction yield of total puerarin flavonoids and polysaccharides were researched by orthogonal experiment design method. Application of the wet method to preparation chewable tablet, filling agent, adhesive and taste masking agent in the best prescription were screened orthogonal experiment design method. According to the Chinese pharmacopoeia, the content of total flavonoids and polysaccharides in chewing tablets were determined using the method of ultraviolet spectrophotometer. Fed rats with high fat, sugar and streptozocin(STZ) to preparation type 2 diabetic model, puerarin chewable tablet role on rats model the fasting blood glucose, blood lipid and antioxidation were researched, respectively.Results:The puerarin flavonoids and its polysaccharide extraction process optimum conditions were at temperature 90?, extracting 2 h, solid-liquid ratio 1:35. The puerarin flavonoids and polysaccharide yield were 10.73% and 10.20%, respectively. The best prescription were mainly puerarin extract 66%, wheat bran 33%, 5%povidone aqueous solution(PVP) 10%, aspartame 0.75%, citric acid 0.5%. With 90% ethanol as moistening agent in dried particles, quantity 1% magnesium stearate as lubricant to preparation chewable tablet. The average weight of chewable tablet was 0.68 g, and the weight difference of inspection. Total content of flavonoids in the chewable tablet was 12.12%, the polysaccharide content was 9.17%. Puerarin chewable high dose group could significantly reduce ? fasting blood glucose(FBG) of diabetes rats(P ?0.05), total cholesterol(TCH)(P?0.05), triglyceride(TG)(P?0.05), superoxide dismutase(SOD)(P ?0.01), increased malondialdehyde(MDA)(P ?0.05).Conclusion:The extraction process is feasible and stable. In the best prescription process conditions, puerarin chewable tablets are good taste, chewing flakes of smoothsurface. Puerarin chewing tablets role on type 2 diabetic rats with hypoglycemic lipid-lowering effect. The conclusion is useful for the development and application of radix puerariae functional health food.
